摘要
目的 研究南瓜多糖对正常及糖尿病模型小鼠支链氨基酸的影响。方法 南瓜干粉经热水提取 ,Sevag法弃蛋白 ,透析 ,乙醇沉淀 ,沉淀干燥物再经SephadexG - 10 0柱层析纯化 ,即得南瓜多糖精品。用聚丙烯酰胺凝胶电泳及玻璃纤维纸电泳证明为均一体。PP的分子量为 1.6× 10 4 。南瓜多糖精品用酸完全水解 ,经纸层析表明由D -葡萄糖、D -半乳糖、L -阿拉伯糖、木糖和D -葡萄糖醛酸组成。其摩尔比分别为 0 .181:0 .0 83:0 .0 6 9:0 .0 31:0 .175。结果 用南瓜多糖精品灌胃 ,剂量在 2 0 0mg/kg和 5 0 0mg/kg时 ,对正常小鼠、四氧嘧啶糖尿病模型小鼠均有降支链氨基酸作用 ,且呈现一定的量效关系。
Objective In order to investigate the effect of pumpkin polysaccharide(PP) on normal mice and alloxan induced diabetic mice.Methods From the Pumpkin,which grows in Shandong,a polysaccharide was extracted with hot water and purified by deproteinization and removal of lipid. Some chemical and physical properties of the polysaccheride were examined. It was homogeneous as judged by electrophoresis.The PP molecular weight was estimated to be 1.6× 10 4 and acidic heteroglycan composed mainly of D- glucose, D- galactose, L-arabinose,Xylose and D-glucuronic aicd in the molar ratios of 0.181: 0.083:0.069:0.031:0.175 respectively.PP was given by the route of ig at a dose of 200mg/kg and 500mg/kg.Results PP was capable to decrease blood branched chain amino acid and in mormal mice,alloxan induced diabetic model mice,and showed dose-effect relation to a cetain extent.Conclusion PP is a ideal practical food drug for treating branched chain amino acid metabolism.
