摘要
为研究处于分类地位不同的 5种棘阿米巴滋养体对黑色素瘤细胞B16的细胞毒性作用的方式、以及细胞毒性作用差异与其致病性的关系。将 5种不同种类的棘阿米巴滋养体分别与体外培养的黑色素瘤细胞B16混合 ,利用细胞免疫荧光技术和透射电子显微镜观察棘阿米巴滋养体对黑色素瘤细胞B16的影响 ,并运用MTT法检测、比较不同棘阿米巴对肿瘤细胞作用的差异。结果显示 :这 5种棘阿米巴滋养体对黑色素瘤细胞B16均有不同程度的细胞毒性作用 ,其机制是诱导肿瘤细胞发生细胞凋亡 。
The aim of this study was to evaluate the cytotoxic effect (CTE) of different species of Acanthamoeba trophozoites on human melanoma B16 cells and to analyze the relation between cytotoxic effect and pathogenicity. B16 cells were co incubated respectively with five isolates of Acanthamoeba trophozoites( A.lugdunensis、A.quina、A.palestinensis、A.quina A.lugdunensis、A.polyphaga ). The photomicrographs of B16 cells were observed by immunocytofluorescence technique and transmission electron microscopy. The Cytotoxic effect was also quantified using MTT method. Results show that B16 cells can be induced to undergo apoptosis and have a sequence of cardinal morphological features of apoptosis when B16 cells were exposed to these five species of Acanthamoeba . In the MTT method,the occurrence of CTE of A.lugdunesis and A.quina trophozoites on B16 cells was more rapid than that of the other species. There was no significant relation between the cytotoxic activity of Acanthamoeba and their pathogenicity.
