摘要
目的研究旋毛虫肌幼虫排泄分泌蛋白对H446小细胞肺癌的抑制作用。方法试验随机分为3组。A组(实验组):将提取的旋毛虫肌幼虫排泄分泌蛋白用无血清培养基稀释成浓度为0.075、0.15和0.3mg/ml,分别与H446细胞共培养;B组(标准对照):将顺铂稀释成12.8、6.4、3.2、1.6、0.8、0.4和0.2μg/ml,分别与H446细胞共培养;C组(空白对照组):H446细胞培养过程中不作任何处理。采用噻唑蓝比色法(MTT法)检测各组不同时间细胞抑制率;采用原位末端转移酶标记法(TUNEL法)观察细胞凋亡情况。结果 MTT法检测各实验组细胞抑制率均随药物浓度的增高和作用时间的延长而升高,与空白对照组比较差异有统计学意义(P<0.01);TUNEL法检测A、B、C组细胞凋亡率分别为85%、43%和2%,差异有统计学意义(P<0.01);A组与B组比较差异有统计学意义(P<0.01)。结论旋毛虫肌幼虫排泄分泌蛋白对H446细胞有抑制作用,可能是潜在的抗小细胞肺癌的生物制剂。
Objective This study monitored the inhibitory action of proteins secreted by Trichinella spiralis on H446 small cell lung cancer to identify the antitumor activity of those proteins in the H446 cell line of small cell lung cancer. Methods H446 cells were randomly divided into three groups: an experimental group (Group A), a standard control group (Group B), and a control group (Group C). In Group A, extracted secretory proteins of T. spiralis were diluted with serum free medium and then added to cultured H446 cells at a concentration of 0. 075 mg/ml, 0.15 mg/ml, and 0.3 mg/ml. In Group B, cultured H446 cells were treated with diluted cisplatin at a concentration of 12.8μg/ml, 6.4μg/ml, 3.2 μg/ml, 1.6μg/ml, 0.8 μg/ml, 0.4μg/ml, and 0.2μg/ml. In Group C, cultured H446 cells were not treated. An MTT colorimetric assay was used to determine the rate of inhibition in the groups of cells at different times. TUNEL was used to observe apoptosis. Results The rate of inhibition was found to increase over time and as the concentration of the added treatment increased. TUNEL revealed that the rate of cell apoptosis was 85 %, 43 %, and 2 %. Few apoptotic cells were noted in Group C, and the difference in the number of apoptotic cells was statistically significant (P〈0.01). Differences in the number of apoptotic cells in Group A and Group B were statistically significant. Conclusion Secretory proteins of T. spiralis successfully inhibited small cell lung cancer in H446 cells in vitro. This suggests that secretory proteins of T. spiralis are a potential biological agent to combat small cell lung cancer.
