摘要
目的观察肺瘤平膏对于Lewis肺癌荷瘤小鼠免疫功能的影响。方法通过流式细胞术测定不同组别肺癌荷瘤小鼠脾脏的DC数量、巨噬细胞数量及表型。结果 DC方面:空白组中DC数量未见明显变化;模型组中DC数量由4.26%逐渐降低到2.59%;肺瘤平膏组在14、21、28 d分别为5.39%、4.83%、4.15%,28 d时,与模型组相比较,2组对比差异具有统计学意义(P<0.05)。巨噬细胞方面:在模型组中,M1型巨噬细胞数量由8.59%逐渐上升到25.98%,同时M2型细胞数量由0.92%逐渐上升到5.42%,M1/M2比值由9.34下降至4.79;肺瘤平膏组中,在14、21 d时,M1、M2型巨噬细胞数量均较模型组增多,21 d时,分别为23.16%及2.77%,其数值相对更高,而在14、21、28 d时,M1/M2比值分别为7.86、8.36、9.62。28 d时,肺瘤平膏组中2种巨噬细胞数量均下降,但M1/M2数值也保持了增高的趋势,说明肺瘤平膏可以促进巨噬细胞向M1型巨噬细胞转化。结论肺瘤平膏可调节荷瘤小鼠脾脏中树突状细胞及巨噬细胞的数量及表达,使其尽可能保持稳定,进而调节荷瘤小鼠的免疫功能处于平衡状态,这可能是其发挥抗肿瘤的作用机制之一。
Objective To observe the effect of Feiliuping Ointment on immune function of Lewis lung cancer bearing mice.Methods Flow cytometry was used to determine the number of DC,macrophage and phenotype in spleen of lung cancer bearing mice of different groups(blank group,Feiliuping Ointment group,model group).Results There was no significant change in the number of DC in the blank group;the number of DC in the model group was decreased from 4.26%to 2.59%;the number of DC in the Lung Ointment group was decreased from 5.39%,4.83%and 4.15%at 14 d,21 d and 28 d,respectively.And at 28 d,there was significant difference compared with the model group(P<0.05).Macrophages:in the model group,the number of M1 macrophages was increased gradually from 8.59%to 25.98%,meanwhile the number of M2 cells increased gradually from 0.92%to 5.42%,and the ratio of M1/M2 decreased from 9.34 to 4.79.In Feiliuping Ointment group,the number of M1 and M2 macrophages was increased more that of the model group at 14 d and 21 d,and at 21 d,they were 23.16%and 2.77%respectively,which were much higher relatively,but at 14 d,21 d and 28 d,M1/M2 ratios were 7.86,8.36 and 9.62,respectively.At 28 d,the number of both kinds of macrophages were decreased,but the M1/M2 value were also kept increasing,indicating that Feiliuping Ointment could promote the transformation of macrophages to M1 macrophages.Conclusion Feiliuping Ointment can regulate the number and expression of dendritic cells and macrophages in the spleen of tumor-bearing mice to make them as stable as possible,and then regulate the immune function of tumor-bearing mice in a balanced state,which may be one of its anti-tumor mechanisms.
