电针对Walker256乳腺癌细胞致骨癌痛大鼠痛阈和脾NK细胞的影响 被引量:6

摘要 目的:以吗啡为对照,观察电针对Walker 256乳腺癌细胞致骨癌痛大鼠痛阈和脾NK细胞的影响,初步电针抗骨癌痛潜在作用优势。方法:实验大鼠随机分为正常组、假模型组、模型组、电针组和吗啡组5组,采用大鼠左侧胫骨腔内注射Walker 256乳腺癌细胞建立骨癌痛模型。电针组给予电针治疗,双侧"足三里"和"昆仑"穴,频率2/100 Hz,强度0.5-1.0-1.5 mA(各10 min),每次治疗30 min,隔日1次。吗啡组大鼠以10 mg/kg的量腹腔注射盐酸吗啡注射液,隔日1次。检测各组大鼠造模前和造模后6、10、16、20 d的体质量和缩腿潜伏期;检测大鼠脾NK细胞活性和百分率(CD_3-CD^+_(161))情况。结果:与正常组比较,造模后10 d和16 d时模型组和吗啡组大鼠体质量增长率降低(P<0.05)。与正常组和假模型组比较,模型组于造模后所有时点缩腿潜伏期缩短(P<0.05,P<0.01);与模型组比较,电针组和吗啡组缩腿潜伏期延长(P<0.01),但电针组短于同期吗啡组(P<0.01)。与正常组和假模型组比较,模型组和吗啡组大鼠脾NK细胞活性降低(P<0.01);电针组大鼠脾NK细胞活性低于正常组大鼠(P<0.05),但高于吗啡组(P<0.05)。与模型组比较,吗啡组大鼠脾NK细胞百分率略增多(P<0.05)。结论:电针治疗大鼠骨癌痛具有镇痛和增强免疫双重作用优势,后者主要是通过其增强骨癌痛大鼠脾NK细胞杀伤活性,而增加非NK细胞数量实现。 Objective: To observe effect of electroacupuncture(EA)on pain thresholds and splenic NK cells in rats with bone cancer pain induced by Walker 256 mammary gland carcinoma cellsand further explore the potential advantage of EA in comparison with morphine.Methods: All rats were divided randomly into normal group, sham model group, model group, EA group and morphine group.The bone cancer pain rat models were established by inoculation of Walker 256 mammary gland carcinoma cells into left tibial cavity.EA(2/100 Hz, 1-1.5-2 mA for 10 min each)was applied to bilateral 'Zusanli'(ST36)and 'Kunlun'(BL60)for 30 min, given once in every other day.Rats in the morphine group were injected intraperitoneally with 10 mg/kg morphine hydrochloride solution, given once in every other day.Body weights and paw withdrawal latencies before and 6 d, 10 d, 16 d, 20 d after modeling were measured.Activity and percentage of splenic NK cells were detected.Results: Compared with the normal group, weight growth rate in the model and morphine group decreased at 10 d and 16 d after modeling(P<0.05).Compared with the normal and sham model group, paw withdrawal latencies in the model group shortened at all time-points we observed(P<0.05, P<0.01).Compared with the model group, paw withdrawal latencies in the EA and morphine group increased(P<0.01), however, paw withdrawal latencies in the EA group were shorter than those of morphine group(P<0.01).Compared with the normal and sham model group, splenic NK cell activity in the model and morphine group decreased(P<0.01).Splenic NK cell activity in the EA group was lower than that of normal group(P<0.05)and higher than that of morphine group(P<0.05).The percentage of splenic NK cells in the morphine group increased slightly in comparison with that of the model group(P<0.05).Conclusions: EA has dual therapeutic effect of analgesia and immune-enhancement in treatment of bone cancer pain.Increasing splenic NK cells activity rather than its percentage might contribute to EA's immunomodulation.
