摘要
目的近年来研究显示,长链非编码RNA(LncRNA)与恶性肿瘤的发生、发展过程密切相关。关于LncRNA与直肠癌的关系目前尚未完全阐明。本研究拟探讨LncRNA AC010145.4在直肠癌组织中表达及临床意义。方法四川省医学科学院·四川省人民医院2012-01-01-2016-12-30收治的直肠癌患者122例、原位癌组织20例、非典型增生组织30例及32例正常直肠组织,通过QRT-PCR法检测LncRNA AC010145.4在正常-非典型增生-癌旁组织-原位癌-结肠癌组织中的表达,分析其表达及临床意义。结果与癌旁组织(1.310±0.092),非典型增生组织(1.550±0.087)及正常直肠组织(1.215±0.143)比较,LncRNA AC010145.4原位癌组织中(2.770±0.650)表达升高,差异有统计学意义,F=29.15,P=0.001。与原位癌及癌旁组织比较,LncRNA AC010145.4在直肠癌组织中表达(8.786±0.335)增高,差异有统计学意义,t=17.730,P<0.001。LncRNA AC010145.4表达与疾病分期(χ~2=15.462,P=0.001)、淋巴结转移(χ~2=4.928,P=0.026)、远处转移(χ~2=4.439,P=0.035)、肿瘤分化(χ~2=14.326,P<0.001)、血清CEA水平(χ~2=10.244,P=0.001)及患者的生存状态(χ~2=42.911,P=0.001)相关。此外,高表达LncRNA AC010145.4患者的无进展生存时间[(13.17±1.36)个月]较低表达者[(27.85±1.23)个月]缩短,差异有统计学意义,χ~2=53.96,P<0.001;低表达LncRNA AC010145.4患者的总生存时间[(43.19±1.84)个月]较高表达者[(26.68±2.02)个月]明显延长,差异有统计学意义,χ~2=29.78,P<0.001。多因素分析显示,LncRNA AC010145.4表达是直肠癌独立预后因素,HR=2.850,P<0.001。结论 LncRNA AC010145.4在直肠癌组织中高表达,可能是直肠癌潜在的预后评估分子标志物。
OBJECTIVE Recent studies have shown that long non-coding RNA(LncRNA)is closely related to the occurrence and development of malignant tumors.However,the relationship between LncRNA with rectal cancer has not been fully elucidated.The purpose of this study was to investigate the expression and clinical significance of LncRNA AC010145.4in rectal cancer.METHODS To tally 122 cases of rectal cancer,20 cases of carcinoma in situ,30 cases of atypical hyperplasia and 30 cases of normal rectal tissues were collected in Sichuan Academy of Medical Sciences and Sichuan People’s Hospital from January 2012 to December 2016.The expression of LncRNA AC010145.4was detected by QRT-PCR.The clinical significance of LncRNA AC010145.4with prognosis of the patients were analyzed.RESULTS The expression of LncRNA AC010145.4in carcinoma in situ tissues(2.770±0.650)was significantly increased than paracarcinoma tissue(1.310±0.092),atypical hyperplasia(1.550±0.087)and normal rectal tissues(1.215±0.143),F=29.15,P <0.001.Compared with paracarcinoma tissue and carcinoma in situ tissues,the expression of LncRNA AC010145.4in rectal cancer tissues(8.786±0.335)was significantly increased,F=17.30,P<0.001.The expression of LncRNA AC010145.4was significantly related to disease staging(χ^2=15.462,P=0.001),lymph node metastasis(χ^2=4.928,P=0.026),distant metastasis(χ^2=4.439,P=0.035),tumor differentiation(χ^2=14.326,P<0.001),serum CEA level(χ^2=10.244,P=0.001)and the patient’s survival state(χ^2=42.911,P=0.001).In addition,the progression free survival time of patients with high expression of LncRNA AC010145.4[(13.17±1.36)months]was shorter than that of the lower expression[(27.85±1.23)months],χ^2=53.96,P<0.001.The overall survival time of the patients with low expression of LncRNA AC010145.4 [(43.19±1.84)months]was significantly longer than high expression patients[(26.68±2.02)months],χ^2=29.78,P<0.001.The expression of LncRNA AC010145.4was an independent prognostic factor for rectal cancer(HR=2.850,P<0.001).CONCLUSION The high ex
