摘要
目的研究淡色库蚊越冬蚊体内海藻糖和海藻糖酶含量的动态变化。方法利用ELISA法测定不同越冬时期淡色库蚊雌成蚊体内海藻糖和海藻糖酶的含量。结果越冬期淡色库蚊滞育蚊所占百分比先增加后减少,越冬中期淡色库蚊全部为滞育蚊。淡色库蚊体内海藻糖含量先增加后降低,到1月份时达最大值,为(225.77±7.461)ng/ml,3月份降至最小值(132.41±2.37)ng/ml。不同越冬时期越冬蚊体内海藻糖含量差异有统计学意义(F=53.59,df=150,P<0.05);海藻糖酶含量先降低后增加,至1月份达最大值,为(9.32±0.23)ng/ml,到越冬末期含量持续减少。不同越冬期淡色库蚊体内海藻糖酶含量差异有统计学意义(F=84.21,df=150,P<0.05)。结论越冬淡色库蚊体内海藻糖和海藻糖酶含量变化较大,在气温最低的1月份含量达最大值,由此推断,海藻糖和海藻糖酶在淡色库蚊越冬过程中起重要作用。
Objective To ascertain dynamic changes in trehalose and trehalase levels in Culex pipiens pallens.Meth-ods Trehalose and trehalase levels in female adult mosquitoes at different overwintering periods were detected using an enzyme-linked immunosorbent assay.Results As overwintering time went on,the percentage of diapausing mosquitoes was increased at first and then decreased,and in mid-overwintering,all mosquitoes were found to be in a state of diapause.The trehalose level in adults increased at first and then decreased;it peaked in January (225.77±7.461ng/ml) and reached its trough in March (132.41±2.37ng/ml).The trehalose level in overwintering mosquitoes differed significantly during different overwintering periods (F=53.59,d f=95,P=0.00).The trehalase level decreased at first and then increased;it peaked in January (9.32±0.23ng/ml)and continued to decrease at the end of overwintering.The trehalase level in mosquitoes differed significantly during different overwintering periods (F=84.21,df=95,P=0.000). Conclusion Trehalose and trehalase levels in overwintering mosquitoes vary greatly and peak in January when the temperature is lowest.Therefore,trehalose and trehalase play important roles in overwintering mosquitoes.
