摘要
柞蚕(Antheraea pernyi)是鳞翅目大蚕蛾科绢丝昆虫,具有可观的经济价值和食用价值。目前,在柞蚕基因组研究中仍没有SNP标记开发及遗传图谱构建的相关报道。以高通量测序为基础的简化基因组测序技术RAD-seq,以其流程简单,不受有无参考基因组的限制,费用低廉,一次测序就可获得海量SNP标记等特点,适用于柞蚕的基因组学研究。RAD-seq技术在许多物种中,已成功用于超高密度遗传图谱的构建、重要性状基因的精细定位等研究。本文主要介绍RAD-seq技术及其在基因组学研究中的应用,为促进该技术在柞蚕遗传标记开发和遗传图谱构建中的应用,推动柞蚕育种工作的快速发展奠定理论基础。
Tussah,Antheraea pernyi,is a kind of silk-spinning insect in Saturniidae of Lepidoptera that has economic value and edible value. At present,there is still no report on single nucleotide polymorphism( SNP) exploitation and genetic map construction in genome research of tussah. Based on high-throughput sequencing,restriction-site associated DNA sequencing( RAD-seq) is appropriate for tussah genome research owing to its characteristics such as simple procedure,no limitation on reference genome,reduced expense,and a large amounts of SNP markers being obtained from one-time sequencing. In many species,RAD-seq has been successfully used in construction of high density genetic map and fine mapping of important characters and so on. Here,we introduce RAD-seq and its application in genome research. This may lay a theoretical foundation on facilitating application of this technique in tussah genetic marker development and genetic map construction,and promoting rapid development of tussah breeding.
