摘要
为了解我国新出现的基因Ⅻ型新城疫病毒(NDV)的分子特性,分析其基因组遗传演化特征,对2010―2012年我国首次分离到的3株基因Ⅻ型新城疫代表株进行基因组测序和生物信息学分析。结果显示,3株病毒F蛋白裂解位点相同为112 RRQKR/F117,符合强毒株特征。病毒基因组长度均为15 192nt,与早期基因型(Ⅰ、Ⅱ、Ⅲ、Ⅳ型)毒株相比,其在NP基因5′端非编码区有6个核苷酸插入。3株病毒在F蛋白信号肽、七肽重复区和跨膜区,以及HN蛋白跨膜区、中和抗原表位等功能区有多处氨基酸变异。病毒基因组遗传进化分析结果显示,3株病毒均属于基因Ⅻ型,但与南美地区分离株不同(基因Ⅻa亚型),我国分离株为基因Ⅻb亚型。
In order to study the molecular characteristics and genetic evolution features of the emerged genotypeⅫ Newcastle disease virus(NDV)in China,the genome sequencing and bioinformatics analysis of 3 representative genotype Ⅻ NDVs isolated during 2010 to 2012 were carried out.The sequence analysis results showed that 3 isolates had the same motif of 112 RRQKR/F117 at the cleavage site of the fusion protein,which was typical of velogenic NDVs.The complete genome lengths of three representative strains were 15 192 nucleotides(nt),having a 6 nt insertion in the5′non-coding region of NP gene when compared with the early genotypes of NDV(genotypeⅠ,Ⅱ,Ⅲ and Ⅳ).Several mutations were identified in the signal peptide,heptad repeat region and transmembrane domain of F protein,and the transmembrane domain and neutralizing epitopes of HN protein.The result of phylogenetic analysis showed the three isolates all belonged to sub-genotypeⅫb,which were different from the viruses isolated from South America(sub-genotypeⅫa).
