摘要
目的:研究大黄不同炮制品配伍后的大承气汤对其泻下作用的变化及其共性关系。方法:将168只小鼠随机分为空白对照组、阳性对照组及大黄不同炮制品配伍后的大承气汤组(6、10g/kg),按容积20mL/kg的剂量给药后,分别用代谢笼积分法、炭末推进作用及Na+-K+-ATPase活性实验,比较大黄不同炮制品配伍后的大承气汤对小鼠的正常泻下作用、炭末推进率及Na+-K+-ATPase活性的差异。结果:大黄不同炮制品配伍后的大承气汤的泻下作用都有不同程度的变化,与空白对照组比较,大黄不同炮制品的大承气汤6g/kg和10g/kg剂量组都能显著增加正常小鼠泻下作用、提高炭末推进率及抑制Na+-K+-ATPase活性;与生大黄大承气汤组比较,熟大黄大承气汤、酒大黄大承气汤、醋大黄大承气汤和大黄炭大承气汤的泻下作用、炭末推进率及Na+-K+-ATPase活性有显著性差异(P<0.05,P<0.01)。结论:大黄不同炮制品的大承气汤泻下作用有差异性变化,这可能与配伍中大黄不同炮制品的主要化学成分引起大承气汤复方中蒽醌类化合物的溶出量变化及大黄不同炮制品药对间的相使相须作用有关。
Objective:To study the purgative activity changes and common relations of Dachengqi Decoction after its compatibility with different processed products of Radix et Rhizoma Rhei.Methods:One hundred and sixty-eight experimental animals were randomly divided into control group,positive control group,and Dachengqi Decoction groups with different processed products compatibility of Radix et Rhizoma Rhei (6g/kg and 10g/kg)according to the volume of 20mL/kg dose. Metabolic cage integral method,carbon powder propelling effect and Na^+-K^+-ATPase activity test were used to compare the normal diarrhea effect,carbon powder propelling rate and Na^+-K^+-ATPase activity of Dachengqi Decoction after its compatibility with differently processed products of Radix et Rhizoma Rhei in mice,respectively.Results:The purgative activity of Dachengqi Decoction after its compatibility of differently processed products of Radix et Rhizoma Rhei has changed in different extent. Compared with the blank control group,Dachengqi Decoction of different processed products of Radix et Rhizoma Rhei at doses of 6g/kg and 10g/kg could significantly increase the diarrhea effect of normal mice,increase the propelling rate of charcoal powder and inhibit the activity of Na^+-K^+-ATPase activity,indicating that the diarrhea effect was significantly different;compared with the Dachengqi Decoction group of raw Radix et Rhizoma Rhei,there were significant differences in the diarrhea effect,the propelling rate of charcoal powder and the activity of Na^+-K^+-ATPase activity of Dachengqi Decoction of cooked Radix et Rhizoma Rhei, wine Radix et Rhizoma Rhei,vinegar Radix et Rhizoma Rhei and Radix et Rhizoma Rhei charcoal (P<0.05,P<0.01).Conclusion: The diarrhea effect of Dachengqi Decoction from different processed products of Radix et Rhizoma Rhei varies,which may be related to the dissolution of anthraquinones in the compound of Dachengqi Decoction caused by the main chemical constituents of different processed products of Radix et Rhizoma Rhei incompatibility
