焦性没食子酸培养阴道毛滴虫方法的建立及其应用 被引量:5

ESTABLISHMENT AND APPLICATION OF PYROGALLIL ACID METHOD FOR TRICHOMONAS VAGINALIS CULTIVATION

摘要 目的 建立阴道毛滴虫焦性没食子酸培养法 ,并研究其在药物敏感实验中的应用。方法 用焦性没食子酸法 ,在固体培养基中培养阴道毛滴虫 ,用打孔法和滤纸片法研究药物敏感性。结果 焦性没食子酸厌氧培养 1- 2 d,在显微镜下仅可见少数虫体聚集的虫落 ,从培养第 3天开始肉眼可见虫落 ,随培养基琼脂浓度增加 ,虫落数和直径均有所下降 ,加入培养基中的虫数越多 ,虫落直径越小 ,随培养时间的延长 ,虫体死亡率逐渐增多。用打孔法和滤纸片法观察药物杀虫效果 ,随药物浓度的增加 ,抑虫环直径增大。结论 焦性没食子酸厌氧培养中 ,培养基的最佳琼脂浓度为 0 .45 %。在直径 6 cm的培养皿中接种 2 0 0个虫数时 ,至培养第 6天虫落直径最大 ,滴虫可存活 9d。用打孔法观察药物杀虫效果较好。常山杀虫效果强于仙鹤草。 Objectives To establish pyrogallil acid method for Trichomonas vaginalis cultivation and observe trichomonacidal effect by this method. Methods Trichomonas vaginalis was cultivated anaerobically with solid media and pyrogallil acid at 37℃. Studies of drug sensitivity were performed by method of hole and filter paper. Results One two days after inoculation the colonies with a few parasites were observed only in microscope. Isolated colonies were readily visible by the third day with naked eyes. It was noted that the number of this organism and the colony diameters decreased as the concentration of agar increased. And the more the number of the organism, the less the colony diameters. Six days later, the mortality of the parasites was rising apparently. Trichomonacidal effects of metronidazole, Changshan, Xianhecao were observed on solid medium with hole or filter paper at 37℃. The diameter of ring of inhibiting parasite growth enlarged as the concentrations of these drugs increased. Conclusion Zero point forty five percent concentration of agar was prefered in trichomonas cultivation by pyrogallil acid method. The proper munber of the parasites on 6 cm diameter plate was 200. By the sixth day after inoculation the diameter of colonies was maximun. Trichomonas vaginalis cultivated in this method were able to survive nine days. It was better to observe the trichomonacidal effect with hole method. Trichomonacidal effect of 25% Changshan was better than that of 25% Xianhecao.
