摘要
目的 在建立慢性病贫血(ACD)大鼠动物模型的基础上,探讨一氧化氮(NO)与关节炎及贫血的关系。方法 在传统佐剂性关节炎大鼠模型基础上,通过追加注射福氏佐剂两次,增强其免疫反应,从而诱发贫血的发生;检测不同处理组大鼠血清铁(SI)、总铁结合力(TIBC)、转铁蛋白饱和度(TS)及血清NO、一氧化氮合酶(NOS)水平。结果 佐剂性关节炎大鼠经追加福氏佐剂,均出现不同程度的贫血,其SI、TIBC、TS水平下降,清铁蛋白(Fn)水平升高(P<0.05),与ACD铁代谢改变特征一致;应用NOS抑制剂大鼠关节肿胀和贫血程度较ACD大鼠减轻,SI、TIBC、TS升高(P<0.001)。结论 成功建立慢性病贫血大鼠模型,NO在ACD大鼠关节炎及铁代谢紊乱中起重要作用。
Objective To investigate the relationship between nitric oxide (NO) and anemia on the basis of establishing the rat model of anemia of chronic disease(ACD). Methods On the basis of traditional adjuvant arthritis rat model lanemia was induced by injecting Freund adjuvant twice so as to enhance the rats immune response , measure the iron .total iron binding capacity(TIBC), trans-ferrin saturation (TS), ferritin(Fn), nitric oxide (NO) and nitric oxide synthase (NOS) in the serum of rats treated with three different ways. Results Varying degree anemia was induced by injecting the Freund adjuvant in adjuvant arthritis rat ,the level SI, TIBC, TS in the serum decreased in ACD rat significantly as compared with the other groups, the level of liver iron content and Fn elevated , which was consistent with the characteristics of iron metabolism disorder in ACD. The degree of arthritic edema and anemia in the rats treated with NOS inhibitor L-NAME was slighter than that of ACD rats , the level of SI, TIBC, TS elevated ( P <0.001).Conclusion NO plays an important role in the disorder of iron metabolism in the ACD rat by establishing of the model of ACD rat successful-ly.
