摘要
近年来植物愈伤组织无性变异系的诱导和选择,已成为植物细胞工程中十分活跃的领域,快中子、γ射线等电离辐射也已开始被用于愈伤组织抗性变异系的诱导。观察不同剂量电离辐射在诱发植物愈伤组织变异中对细胞结构的影响。
Callus of sainfoin (Onobrychis viciae folia Scop) was radiated by fast neutron at different dosage. Small pieces of callus were fixed, sectioned and observated with EM400 electron microscope at 3 rd, 6 th and 12 th day after radiation. The radiation of fast neutron at low dosage (1. 3×10^(12) n/cm^2) brought about light damage to cells. The nucleus became smaller and the nucleolar structure was not distinet. A number of plastoglobuli appeared in the plastids and the lysosome and other inclusion bodies appeared in the vacuoles. But these damages were repaired in development of cells and then the cells continued to grow, develop and divide normally. The radiation at high dosage (1.56 x 10^(13) n/cm^2) brought about heavy damages to the cells. The nuclear membrane, membrane of endoplasmic reticulum and plastids were disintegrated and disappeared. The chromatin was condensed and vacuoles were fulled with lysosomes and autophagosomes. These damages were not revesible, finally cause disintegrations of nucleus, plastids, endoplasmic reticulum and other organoids in callus cells.
