摘要
本文研究微载体半连续悬浮培养表达乙型肝炎表面抗原(HBsAg)的基因工程哺乳动物细胞(Bu3)的工艺过程,建立了细胞在微载体上的生长模型、葡萄糖消耗和乳酸生成模型。实验表明,微载体悬浮培养系统能使贴壁依赖性Bu3细胞达到高密度和产物高表达。
The process is studied in which Bu3 cells secreting HBsAg are cul-tured on microcarriers semi-colltinuously, a propagating model of Bu3cells on microcarriers, glucose consumption, lactic acid accumulationmodels are established. It is illustrated that microcarrier culture techno-logy enables the anchoraged Bu3 cells to reach high yield and highHBsAg expression.
