摘要
通过根瘤农杆菌 (Agrobacterium tumefaciens) L BA4 4 0 4中 Ti-质粒的介导将含有透明颤菌血红蛋白 (Vitreoscilla Hemoglobin,VHb)基因 vgb和博来霉素 (bleomycin)抗性基因转移到产黄青霉 (Penicilliumchrysogenum)中 ;利用根瘤农杆菌介导转化产黄青霉孢子时需要有乙酰丁香酮诱导 ;转化菌丝体时乙酰丁香酮不是必要的 ,但一定浓度的乙酰丁香酮可提高转化率 ,与原生质体转化法相比可提高转化率 10倍左右。
vgb and bleomycin resistance genes could be efficiently transferred to a filamentous fungi Penicillium chrysogenum by the aid of Ti plasmid of Agrobacterium tumefaciens LAB4404 and the transferred genes were integrated at a chromosomal locus of P.chrysogenum . Transformation could be enhanced in the presence of acetosyringone(AS) when being carried out in the conidia of A.tumefaciens LAB4404. However AS is not necessary for transformation of genes in mycelium. The efficiencies of transformation could be improved up to 10 folds as compared with conventional techniques for transformation of P.chysogenum protopalsts.
