摘要
目的 探讨趋化因子RANTES在狼疮肾炎的表达情况及其对狼疮肾炎肾损伤的作用。方法 复制慢性移植物抗宿主(GVHD)小鼠模型,应用免疫组织化学和逆转录多聚合酶链式反应检测不同时期狼疮样小鼠肾组织RANTES的表达。结果(l)正常小鼠肾组织中未见RANTES表达,8周狼疮样小鼠肾组织中,见部分肾小管上皮细胞表达RANTES;12周狼疮样小鼠肾组织中,大部分肾小管表达强的RANTES,并与小管间质损伤总积分呈正相关关系(r=0.912,P<0.01),肾小球RANTES阳性细胞数与肾小球硬化指数也呈正相关关系(r=0.837,P<0.01);(2)正常小鼠肾组织中未见RANTES mRNA表达,12周狼疮样小鼠肾组织 RANTES mRNA表达明显高于8周狼疮样小鼠,RANTES mRNA/β-actin mRNA有显著性差异(P<0.05) 结论 RANTES在自身免疫性肾炎中起重要作用,是加剧肾损伤的关键因子之一.
Objective To investigate the expression of chemokine RANTES (regulated upon activation normal T cell expressed and secreted) and its role in the pathogenesis of chronic graft-versus-host disease (GVHD) murine lupus nephritis ( LN) model. Methods A microwave-based immunohistochemistry (SP) and RT-PCR were used to detect the expression of RANTES in the kidney of model and normal control. At the same time, the correlation between the expressions of RANTES and renal injures in model was examined. Results At twelfth week the expression of RANTES protein and mRNA in model was higher than that at eighth week and that of normal control( P <0.05). The expression of RANTES protein in renal tissue of model showed a positive correlation with histological changes. Conclusion RANTES. involved in the process of renal injury in vivo, may play an important role in the pathogenesis of LN.
