摘要
目的 :评价前期已构建的含有重组人白细胞介素 12基因的真核载体的表达能力。方法 :重组IL 12真核表达载体体外转染 2 93细胞后 ,收集 72h培养上清 ,用LDH法检测上清对自然杀伤活性的增强作用 ,并用标准品进行对比分析。结果 :实验前期所构建的表达载体可真核表达IL 12蛋白 ,其含量在 2 .5~ 5 .0ng/ml之间。 结论 :可直接测得IL 12的生物活性 ,并能由此对所构建的载体的表达能力进行分析。
Objective: To assess the expression capability of the previously constructed eukaryotic expression vectors carrying the recombinant human interleukin 12(hIL 12) gene. Methods:The recombinant hIL 12 eukaryotic expression vectors were transfected by the 293 cells, and the supernatant was harvested 72 hours after the transfection. The enhanced effect in natural killer activity due to the addition of the post transfection cell supernatant was examined by LDH and was compared with the IL 12 standard sample. Results: The IL 12 eukaroytic expression vectors constructed previously in our laboratory can express the IL 12 protein and its concentration was between 2.5~5.0 ng/ml. Conclusion: This method can be applied to measuring the bioactivity of the IL 12 and thereby assessing the expression capability of the constructed vectors.
