摘要
应用门静脉二步灌流和Percoll不连续密度梯度离心法,从大鼠肝分离获得高纯度的大颗粒淋巴细胞(LGL),细胞获得率为1.0~1.8×10~5个/每克肝组织.经May-Giemsa染色观察,细胞直径10~16μm,核多呈肾形或有切迹,核一侧胞浆较丰富,含大小不同,数量不等的嗜苯胺蓝颗粒,颗粒呈酸性磷酸酶阳性.电镜下,细胞表面有绕毛,核染色质高度浓缩,一侧胞质内有数量不等的溶酶体.以YAC-1细胞株为靶细胞的~3H-TdR释放试验证明,分离的LGL具有自然杀伤活性.肝LGL分离成功为深入研究其免疫特性和抗肝癌作用及其与肝内其他细胞的关系奠定了基础.
The large granular lymphocytes (Pit cell) in high purity were isolated from rat liver by two-step portal vein perfusion method and discontinuous Percoll density centrifugation.The yield of LGLs was 1.0~1.8×10~5 cells per gram of the rat liver.In May-Giemsa stained cell smears,the cells were 10~16μm in diameter,with kidney-shaped nuclei,rich cytoplasm and typical azurophilic granulas.Acid phosphatase was positive.The ultrastructural features of these cells were the characteristic electron-dense granules located in one side of the nucleus.The cytotoxicity assay showed that these cells had NK activity against YAC-1 cells.
