高效液相色谱法测定脑溢安浸膏在大鼠体内大黄酸的含量 被引量:8

摘要 目的 建立大鼠血清大黄酸的高效液相分析法 ,测定不同次数口服给予脑溢安浸膏血清中大黄酸的含量。方法 大鼠单次给 2g脑溢安浸膏和 4d 7次给药 (1g/次 ,2次 /d) ,于不同时间心内取血用HPLC测定大黄酸含量 ;HPLC条件 :C18柱 ,以 0 .1 %磷酸 -甲醇 (3 0∶70 )为流动相 ,紫外检测波长 43 5nm。结果 单次与多次给药均在 1h给药后达到最高血药浓度 ,但单次给药血药浓度迅速下降 ,而多次给药可维持较长时间的血浓平台。大黄酸在 0 .0 3 2~ 4.8mg·L-1范围内呈线性 ,r=0 .999,阴性无干扰 ,精密度RSD为 1 .83 % ,加样回收率为 96.9%。结论 检测方法简便、灵敏、准确、稳定 ;本品以每日 2次 。 Objective: To establish a HPLC method of Rhein in rat serum and determine the concentration of Rhein after oral administration of NaoYiAn extract.Motheds: Rats were given 1g NaoYiAn extract only one time or 7 times in 4 days,blood was taken from heart at different time points,Rhein in these serum was determined by HPlC:C 18 column was used,the mobile phase was consist of a mixture of 0.1% H 3PO 4,methanol (30∶70V/V),the detection wavelength was 435nm.Results:Concentration of Rhein eaches the highest level one hour after administration of NaoYiAn in different doses,but concentration of Rhein decreases rapidly in one dose,that in many doses maintains at high level.A Linearity was obtained over the concentration of 0.032~4.8?mg·L -1 ,r=0.999.The average recovery was 96.9% with RSD=1.83%.Conclusion:The determination of Rhein in rat serum is Stable,simple,sensitive and exact;It is suitable to give the drug two times every day in successive way.
