摘要
取不同基因型的玉米自交系叶片约 0 5cm接种在MS(大量 ) +B5(微量 ) +6 BA(0 5~ 2mg/L) +2 ,4 D(1~ 4mg/L) +IAA (1mg/L)的诱导培养基上 ,结果表明 :玉米叶片愈伤组织及丛生芽的形成能力因其形态部位的不同而呈显著差异 ,从基部向上依次递减。丛生芽的形成与基因型及激素配比有关。通过RAPD分析 ,发现正常再生植株与畸形苗和白化苗的基因组之间存在差异 ,说明在愈伤组织诱导及继代过程中玉米的基因组发生了变异。
The leaves of different genotypes of inbred maize were cultured in induction media, which contained MS (medium's macroelements), B 5 (medium's microelements), 6 BA (0 5~ 2 mg/L ), 2,4 D(1~4 mg/L) and IAA(1 mg/L) After three weeks of induction culture, it was showed that the formation ability of maize leaf segments decrease gradually from basal to upper segments The generation of multiple shoot clumps showed significantly different in different maize genotypes and at different hormone combinations The results of randomly amplified polymorphic DNA(RAPD) showed that the genomic DNA was modified during culture
