摘要
建立适合大黄药材、大黄浸膏及清胃颗粒剂过程分析与质量控制的反相高效液相色谱方法。采用YWG—ODS 10 μm(2 0 0× 4.0mmID)色谱柱 ,流动相组成为甲醇—水—高氯酸 (75∶2 5∶0 1V/V) ,检测波长 2 5 4nm ,流速1.0ml/min ,柱温控制 2 0℃。以芦荟大黄素、大黄素、大黄酸、大黄酚和大黄素甲醚的峰面积作为定量信息 ,标准曲线外标法测定样品含量。结果表明 ,清胃颗粒剂中五种游离蒽醌成分的平均加样回收率分别为芦荟大黄素95 6 1%(RSD3 0 5 %)、大黄素 98 0 3%(RSD2 77%)、大黄酸 10 0 1%(RSD2 2 1%)、大黄酚 97 4 9%(RSD3 6 4%)和大黄素甲醚 96 79%(RSD4 12 %)。本法操作简便、检测灵敏 ,适合于大黄药材、大黄浸膏及清胃颗粒剂过程分析与质量控制。
A RP-HPLC method for the production analysis and quality control in radix rhei, rheum extracts and qingwei granules was established. The RP-HPLC separation was carried out on YWG-ODS colum (200×4.00mm I.D., 10μm) with methanol-water-perchloric acid (75∶25∶0.1V/V) as the mobile phase and UV detection at 254nm. The flow rate was 1.0ml/min and the column temperature was kept at 20℃. The contents of free anthraquinones were determined by means of external standard method. The average recoveries of 95.61%(RSD3.05%) for aloe-emodin, 98.03%(RSD2.77%) for emodin, 100.1%(RSD2.21%) for rhein, 97.49%(RSD3.64%) for chrysophanol and 96.79%(RSD4.12%) for physcion were obtained in rheum Qingwei granules, respectively. The RP-HPLC method is accurate, sensitive, simple and suitable for the production analysis and quality control in radix rhei, rheum extracts and Qingwei granules.
