摘要
目的 研究丹参不同成分对肿瘤细胞和正常细胞生长的影响。方法 采用MTT法测定不同丹参制剂对Bel 740 2细胞和人外周血干细胞生长的影响 ,采用细胞计数和放射免疫法测定不同丹参制剂对Bel 740 2细胞增殖和甲胎蛋白分泌的影响。结果 丹参酮ⅡA 对Bel 740 2细胞和外周血干细胞的IC50 值分别为 ( 5 .96± 2 .46 ) μg/ml和 ( 2 3 .70± 15 .33) μg/ml ;丹参浸膏的IC50 值则为 ( 95 .2 0± 17.38) μg/ml和 ( 14 2 .2 4± 36 .5 1) μg/ml ;丹参注射液对 2种培养细胞的生长均有支持和保护作用。丹参酮ⅡA和丹参浸膏虽可抑制Bel 740 2细胞的增殖 ,但不影响甲胎蛋白的分泌量。结论 丹参酮ⅡA 和丹参浸膏对体外培养的肿瘤细胞和正常细胞均有抑制作用 。
Objective To investigate the effects of three composition of Salviae Miltiorrhizae(SM) on growth of tumor and normal cells.Methods MTT assay was used to test the effect of drugs on growth of Bel 7402 and peripheral blood stem cells.Growth rate and alpha fetoprotein(AFP) value of Bel 7402 cell were examined by cell count and radioimmunoassay methods.ResultsTanshinone Ⅱ A(Tan)?SM extractum inhibited growth of Bel 7402 and peripheral blood stem cells,the IC 50 value of Tan was (5.96±2.46) μg/ml and (23.70±15.33) μg/ml respectively,and the IC 50 of SM extractum was (95.20±17.38) μg/ml and ( 142.24 ±36.5) μg/ml respectively.SM injection supported the growth of two sort of culture cells.The growth rate of Bel 7402 cells were inhibited by Tan and SM extractum,after culturing 4 days,cell counts of drug groups were 46.39% and 50.52% respectively to control group,but AFP value was similar amoung three groups.Conclusion Tan and SM extractum inhibite both tumor and normal cells in vitro,SM injection supports growth of the two sort of cells.
