摘要
目的 探讨鱼棉平喘方对哮喘豚鼠的作用机理。方法 通过平喘实验观察引喘潜伏期及放射免疫分析法测定哮喘豚鼠分组治疗后血浆环磷酸腺苷 (cAMP)、环磷酸鸟苷 (cGMP)水平。结果 测定各组豚鼠给药 1周前、后的引喘潜伏期 ,结果显示本方治疗各组与正常组以及与治疗前各组比较 ,均显著延长引喘时间 (P <0 .0 1) ,与阳性药组比较无显著性差异 (P >0 .0 5 )。将豚鼠造模和复制引喘后分组给药治疗 1周后取血测定各组豚鼠血浆中cAMP、cGMP水平及cAMP/cGMP比值 ,结果显示本方治疗各组与模型组比较均显著升高血浆cAMP水平及cAMP/cGMP比值、降低血浆cGMP水平(P <0 .0 1~ 0 .0 5 )。与阳性药组、正常组比较均无显著性差异 (P >0 .0 5 )。结论 本方有明显的平喘作用 ,其作用机理与其调节机体cAMP。
OBJECTIVE To investigate into the mechanism of the effect of Yumian Pingchuan Prescription on guinea pig with asthma. METHOD The incubation period of asthma induction was observed and the levels of cAMP and cGMP of guinea pigs were determined after treatment with radioimmunoassay. RESULT The incubation periods of asthma induction one week before and after the treatment were determined. The result showed that this prescription could markedly prolong the time of asthma induction ( P <0.01) with no significant difference as compared with the positive drug group ( P >0.05). One week after the treatment following asthma induction with guinea pig models, the levels of cAMP and cGMP in the guinea pig plasma and cAMP/cGMP ratio were measured. The result showed that Yumian Pingchuan Prescription could markedly increase the level of plasma cAMP and cAMP/cGMP ratio, and reduce the level of plasma cGMP ( P <0.01~0.05), with no significant difference as compared with the positive drug group and normal group ( P >0.05). CONCLUSION Yumian Pingchuan Prescription has obvious anti asthma effect and its mechanism is probably due to its function to regulate the body's levels of cAMP and cGMP and stabilize cAMP/cGMP ratio.
