黄瓜根尖微粒体吲哚乙酸结合蛋白的分离纯化与鉴定 被引量:1

摘要 用黄化黄瓜苗根尖为材料,经匀浆、分级离心得到微粒体,然后经丙酮分步抽提得到含有吲哚乙酸(IAA)结合蛋白粗制品,再经IAA-BSA-Sepharose 4B亲和层析,用3mol/L KCNS解脱,得到纯化的IAA结合蛋白。ELISA试验结果表明,该结合蛋白能完全阻断IAA多克隆抗体对IAA-OA的结合反应,并且能竞争抑制IAA多克隆抗体与游离IAA的结合。 Cucumber root tips were used to fractionate microsomal vesicles which were afterwards applied to solubilize IAA binding protein with acetone. Crude solubilized IAA binding protein preparations were then loaded on IAA-BSA-Sepharose 4B column. With ELISA technique eluate by 3 mol/L KCNS would completely block the binding of IAA to anti-IAA antibody, and could also bind with free IAA, which suggested that the protein purified by affinity chromatography be IAA binding protein.
