摘要
目的 探讨眼部鳞状细胞癌中细胞黏附分子变异体 6 (clusterofdifferentiation 44variant 6 ,CD4 4V6)蛋白表达及与增殖细胞核抗原 (proliferatingcellnuclearantigen ,PCNA)的关系。方法应用链霉亲和素 生物素 过氧化物酶复合物免疫组化法检测 35例眼部鳞状细胞癌、2 0例乳头状瘤和11例正常眼睑皮肤组织标本CD4 4V6蛋白表达。结果 CD4 4V6蛋白在眼部鳞状细胞癌 (squamouscellcarcinoma ,SCC)中的阳性表达率为 6 2 9%(2 2 / 35 ) ,而在乳头状瘤的阳性表达率为 15 0 %(3/ 2 0 ) ,在 11例正常眼睑皮肤组织中均无CD4 4V6蛋白的阳性表达 ;恶性组CD4 4V6蛋白的阳性表达率明显高于良性组(χ2 =11 75 7,P <0 0 1)和正常组 (P =0 0 0 0 1) ,8例淋巴转移的CD4 4V6蛋白的阳性表达率明显高于无转移者 (P =0 0 49) ;CD4 4V6蛋白的阳性表达组其PCNA指数明显高于阴性表达组 ,差异有非常显著性(t=2 0 2 1,P <0 0 1)。结论 CD4 4V6分子可能与眼部鳞状细胞癌的恶性表型、浸润及转移有关 ,CD4 4V6与PCNA蛋白的阳性表达呈正相关 ,两者同时检测可能对判定患者预后有临床参考价值。
Objective To investigate the expression of cluster of differentiation 44 variant 6 (CD 44V6) and proliferating cell nuclear antigen (PCNA) in squamous cell carcinomas (SCC) in eyes. Method Streptavidin-biotin peroxidase complex (SABC) immunohistochemical method was used to study the expression of CD 44V6 and PCNA in 35 cases with squamous cell carcinoma in eyes, 20 cases with papilloma, 11 cases with normal eyelid tissue. Results The CD 44V6 positive rate was 62.9% (22/35) in squamous cell carcinomas in eyes, 15.0% (3/20) in papillomas, none in 11 cases with normal eyelid tissue. The positive expression rate of CD 44V6 in the malignant group was significantly higher than that in the benign and control group, and the positive expression rate of CD 44V6 in cases with metastasis was significantly higher than that without metastasis. PCNA labeling index (PI) in the CD 44V6 positive expression of tumors was significantly higher than that in the CD 44V6 negative expression of tumors (t=20.21,P<0.01). Conclusions The over-expression of CD 44V6 is correlated with the progress and metastasis of SCC. CD 44V6 protein expression is correlated positively with PCNA expression, and the evaluation of both CD 44V6 and PCNA is useful for predicting the prognosis.
