硬头鳟(Oncorhynchus mykiss)早期发育过程中红血细胞核的尺度变化和类型转换 被引量:3

摘要 硬头鳟早期发育过程中存在三种红血细胞核类型,圆形的幼体核(L核,在圆盘形的红血细胞内),椭圆形的成体核(A核,在椭圆盘形的红血细胞内)和近椭圆形的未成熟的成体核(ImA核,在近椭圆盘形的红血细胞内)。L核从胚胎早期发生(眼点期前后)至卵黄吸收完毕后(受精后65日,孵出后33日)消失,其间核直径逐渐减少,减幅达29.7%。A核从胚胎孵出时开始出现,至卵黄吸收完毕后,A核基本取代其它类型核,其间核长短径持续发生变化。长径逐渐增加,短径逐渐减少,这种变化直至受精后99日才稳定下来,此时A核长径增幅为43.1%.短径减幅为22.3%.长短径比率由1.11增至2.04。ImA核在胚胎孵出时开始出现,但比例很少,尺寸变化不大,在卵黄吸收完毕稍后(受精后74日,孵出后42日.)可忽略大计(基本上转变为A核)。在胚胎孵出前,红血细胞核全为L核;在卵黄吸收期间,L,A和ImA3种混合存在,核大小及形状发生急剧变化,核类型迅速发生转换,最终全为A核所取代.这种变化和Ichiro(1974)所描述的虹鳟红血细胞的变化基本一致。在印黄吸收完毕后(受精后74日),A核尺度相对稳定。Wright染色后在显微镜下观察,细胞核着色深,核形? Yhere are tree types of erythrocyte nuclei exists in the early developing Steelhead trout,larval type (nucleus L. in the round disc-like erythrocyte),abuly type (nucleus A,in the elliptic disc-like adnly erythrocyte)abd immature adult type(nucleus ImA,in the immature erythrocyte). The round nucleus L ap=pears in the early embryonic development and remaims until swim-up sathe (65 days post-fertilization). They decrease gradually in diameter by 29. 7%. Tthe elliptic nucleus A appears from hatching(33 days post-fertilization) and dominate after swim-up stage. They coneynuously increase in majoraxis (a)by 43. 1 %and decreases in minor axis (b)22. 3% until 33 days post-swim-up (98 days postfertiization). The ratio a/bincreases accoriengly fron 1. 11to 2. 04. The nucleus ImA is presusde to be the precursor of nucleus A. they appear from hatching with a small proprtion.vary slightly,peak in numbers prior to swim-up and transit totally to nucleus A10days post - swim - up.Erythrocyte nuclei are larval type at prehatching. During yolk absortion. the three nucoear types esixt simulta-neously with a dramatic size variation and type transition. These changes keep until swim-up stage when if is dominated by the tytical elliptic abult erythrocyte nuclei, in respomdency fundermentally to the transition of erythrocytes im Rainbow trout described by Ichiro. Afterwatds nuclei A keep relativelt unchaigedin sizeabd can be deep -with Wright stain that can facilitate erythrocyte nuclear measurement for ploid determination. It is therefore, srggested to take sanples post -swim-up during the earlyde -velopment in Salmonids,such ac Steelhead Tuout.
