摘要
目的 建立MBP特异性自身反应性T细胞克隆 ,为进一步探讨EAE的发病机制和治疗奠定基础。方法 以MBP肽免疫C5 7BL 6小鼠 ,取其淋巴结细胞 ,加同系鼠脾细胞体外共同培养于含IL 2的培养液中 ,并以MBP肽反复刺激。收集培养细胞 ,以流式细胞仪进行鉴定。结果 流式细胞仪分析显示 ,90 %以上的细胞为CD4+ T细胞。结论 成功地建立了C5 7BL 6小鼠的MBP反应性T细胞克隆 ,该细胞克隆为MBP特异性自身反应性CD4+ T细胞。
Objective To establish myelin basic protein (MBP) autoreactive T cell line for investigating the pathogen mechanism and treatment method of EAE. Methods Lymphocytes were taken from lymph nodes of C57BL/6 mice immunized with MBP peptide 68 86 and cultured in vitro with syngenic splenocytes in medium which IL 2 and MBPp68 86 was added into.Some days later, the cells were collected and identified with FACS. Results The result showed that more than 90% of the cells were MBP specific CD4 + T cells. Conclusion MBP specific T cell line which belonged to CD4 + subset is successfully established from C57BL/6 mice.
