摘要 目的 为了有效收集一次随机尿液中微量蛋白质 ,以作为职业性肺癌及其他肿瘤患者尿微量基因蛋白研究的基础。方法 本实验应用低温加压超滤系统对肺癌及健康者尿微量蛋白质进行超浓缩 ,并对尿微量蛋白的优化收集条件进行探讨。结果 显示肺癌及健康者 10 0ml尿液经超浓缩和纯化后 ,可得到尿微量蛋白 610mg ,并发现经冰冻保存超过 1d的尿样蛋白明显下降。结论 提示尿样在 4℃的保存条件下进行超浓缩和纯化处理后 ,可得到很高的尿微量蛋白质。能满足多次重复试验。 Objective To collect microprotein effectively in urine for finding biomarker of occupational lung cancer. Methods Urinary microprotein from lung cancer patients were measured with different conditions. Results The higher levels of microprotein in urine were found in specimens concentrated with special filter system at 4℃, up to 6 100 g/L. However, the level was shown to be decreased after freezing preservation more than one day. Conclusion The method can serve as a strong base for study on micro oncoprotein in urine from various cancer patients.
