摘要
目的 研究经门静脉途径 (脾脏 )注射131I标记抗CEA单抗后对荷瘤鼠血清、腹水和瘤组织中CEA水平的影响。方法在接种结肠癌细胞悬液后 1 0min内 ,分别经脾脏和经尾静脉注射131I标记抗CEA单抗 ,对照组注射131I标记正常小鼠IgG。结果 经脾RAIT组荷瘤鼠血清、腹水和瘤组织中的CEA水平分别为 ( 2 3.8± 4 .3)、( 4 0 .1± 5.2 )、( 4 5.7± 7.0 ) μg/L ,对照组则为 ( 4 4.1±4 .7) ,( 52 .3± 7.0 ) ,( 6 7.0± 9.2 ) μg/L ,差异均有非常显著性 (P <0 .0 1 ) ;经脾RAIT组荷瘤鼠血清CEA水平也低于经尾静脉RAIT组 ( 2 9.7± 4 .8μg/L) ,两组之间具有显著性差异 (P <0 .0 5)。 结论 经门静脉途径注射131I标记抗CEA单抗较经尾静脉注射能更有效地降低荷瘤鼠血清中的CEA水平。
Objective To study the changes of CEA level in serum, ascites and liver metastatic tumor after treated with 131 I labeled anti CEA McAb injected intrasplenically in nude mice. Methods Ten minutes after injection of colonic cancer cells into spleen, 300mCi 131 I labeled anti CEA McAb was injected through the splenic or the tail vein, respectively. 300mCi 131 I labeled anti IgG was injected through the tail vein in control group. Results Four weeks later, the levels of CEA in blood, ascites and metastatic tumor were 23.8±4.3, 40.1±5.2, 45.7±7.0 (μg/L) in intrasplenic RAIT group, and 44.1±4.7, 52.3±7.0, 67.0±9.2 (μg/L) in control group, respectively ( P <0.01). The serum CEA level in intrasplenic RAIT group was less than that in intratail vein RAIT group ( P <0.05). Conclusion Injection of 131 I labeled anti CEA McAb can lower the serum CEA level more obviously through the portal vein than through the tail vein.
