摘要
目的 采用化学交联方法将α-糜蛋白酶固定化于胶原蛋白凝胶。方法 以戊二醛促进提取的I型胶原蛋白与α -糜蛋白酶形成西夫碱 ,从而生成固定化糜蛋白酶 ,并计算固定化效率 ,观察 pH、温度对酶活性影响及固定化对胶原蛋白生物学特性的影响。结果 作出pH、温度等外界因素对固定化酶活性的影响曲线 ,并得到具一定活性的固定化α-糜蛋白酶。结论 已成功地制备以胶原固定化α -糜蛋白酶的创面外用膜片 ,可作进一步研究。
Objective To immobilize α- chymotrypsin on collagen gel using chemical linking.Methods Type I collagen extracted and α- chymotrypsin were enhanced by glutaric dialdedyde to form schiffis alkali, which produced immobilized α-chymotrypsin. The efficiency of immobilization was calculated. The effects of pH and temperature on the activities of α- chymotrypsin, and that of immobilization on the biological qualities of the collagen were observed.Results The effect curves of pH and temperature were drawn and immobilized α-chymotrypsin which had some activities were created.Conclusion In the study,an external used membrane for wounds is successfully prepared with collagen gel-immobilized-α- chymotrypsin ,which needs further study
