摘要
为探讨氯化消毒副产物二溴乙酸的致癌性及其可能的致癌机制,使用NIH3T3细胞进行 了恶性转化试验,并用刀豆蛋白A凝集试验验证细胞恶性特征,用流式细胞仪对转化后细胞进 行了细胞周期时相分析.结果表明,二溴乙酸能导致细胞恶性转化,转化细胞能引起低浓度刀 豆蛋白A凝集,细胞周期出现G2/M期阻滞.可见二溴乙酸具有潜在致癌性,有必要对其进行进一 步研究.
NIH3T3 cell malignant transformation experiment was used to explore the carcinogenic activity of a by-product of chlovinating disinfection-bromoacetic acid(DBA) and its possible mechanism. And the malignant character of transformed cell was tested and verified by ConA agglutination. Cell circle of transformed cells was analyzed by flow cytoinstrument. The results showed that DBA could induce the malignant transformation of NIH3T3 cell. The transformed cell could cause the agglutination of low concentrated ConA and arrest of cell circle in G2/M stage. It is thus evident that DBA possesses potential carcinogenic ability and it is necessary to study it further.
