摘要
在枯草杆菌中 ,嘌呤核苷酸合成途径中存在AMP和GMP转变成IMP的环行途径 ,而在该菌的黄嘌呤缺陷型菌株中IMP至XMP的分枝途径被阻塞 ,同时在该菌株内又有强的 5’ -核苷酸酶活性 ,这使其可以生成腺苷 (AR) ,但反馈抑制是腺苷产率的一个重要影响因素。使用季铵盐阳离子表面活性剂十六烷基三甲基溴化铵 (CTAB)可改善细胞的渗透性 ,减少细胞内产物的反馈抑制 ,促进产物的分泌。结果表明 ,在发酵 60h添加 4%的CTAB能使产率从 3.0 g/l提高到3.6g/l,提高幅度达 2 0 %。
There were two circular ways in synthesis of purine nucleotide, caused by both AMP and GMP returning to IMP in B.subtilis . But in the strain(X ), the branch way from IMP to XMP was blocked. At the same time 5' nuclease strongly existed in the strain ,which made it possible to produce adenosine. If the feedback inhibition which intensely influenced the adenosine productivity was relieved, it was feasible to produce adenosine in the strain(X ). The hexadecyl trimethlammonium bromide(CTAB) was used for improving the permeability of cell to promote secrete of adenosine. As a result, the lower concentration of production in cells would relieve the feedback inhibition. The addition of 4% CTAB at the sixtith hour of fermentation would promote productivity from 3.0g/l to 3.6g/l with 20% of increasement.
