摘要
油酸(OA)注入大鼠离休肺动脉后出现损伤,表现为丙二醛、乳酸脱氢酶增高、超氧化物歧化酶、过氧化氢酶下降。经用钨及别嘌呤醇后,上述变化减轻,肺组织黄嘌呤氧化酶活性降低,过氧化氢酶活性受到保护,提示在缺乏血液成分时,OA所致肺损伤,其氧自由基主要来自肺血管内皮细胞的黄嘌呤氧化酶。活性氧中过氧化氢具有重要意义。
The role of oxygen free radicals (OFRs) in pathogenesis and effect of tungsten and allopurinol on oleic acid (OA) induced isolated rat lung injury was investigated; The rats were divided into 4 groups: (1)Control?;(2) OA( 8 uI); (3) All-opurinol +OA;(4) Tungsten+OA ( pretreatment with tungsten rich diet for 3 -4 weeks). The results were as follows: (1) Compared with the control group, the content of malondiadehyde(MDA) and lactate dehydrogenase (LDH) increased, and the superoxide dismutase (SOD) activity decreased in OA group. The lung coefficient signi ficantly correlated with the content of MDA and LDH, and negatively correlated with the SOD activity. (2) The OA induced lung injury attenuated in group 3 and 4.Compared with OA group, the catalase activi ty and tungsten/molybdenum ratio increased and the content of molybdenum and xanthine oxnthine oxidase activi ty decreased in the lung tissue of tungsten + OA group. The lung coefficient significantly correlated with the xanthine oxidase activity. It is suggested that:(1) OFRs play a key role in the development of OA induced lung injury and the hydrogen peroxide might contribute to it; (2) In absence of blood components the OFRs mainly derived from xanthine oxidase in the endo-thelial cells.
