摘要
建立了高效液相色谱法检测人胃黏膜组织中的左氧氟沙星.共收集我院门诊37例幽门螺杆菌(Helicobacter pylori,Hp)阳性的慢性胃炎患者,用含左氧氟沙星三联方案(埃索美拉唑20 mg+阿莫西林1 000 mg,2次/天,左氧氟沙星500 mg,1次/天)根除Hp,疗程7d.受试者于疗程结束后接受胃镜检查,胃窦部取材3块并制成匀浆,利用HPLC法检测人胃黏膜组织中的左氧氟沙星.采用Hypersil ODS C18色谱柱,流动相为乙腈∶50 mmol/L枸橼酸溶液∶1 mol/L乙酸铵溶液(27∶72∶1),检测波长295 nm.人胃黏膜组织中左氧氟沙星在0.5~10 μg/g范围内线性关系良好,低、中、高浓度质控样品的方法回收率分别为96.4%、95.2%和102.8%;日内、日间RSD分别为6.70%、2.01%、1.46%和7.40%、4.56%、3.62%.37例患者胃黏膜中左氧氟沙星浓度为(6.05士2.47) μg/g.
An HPLC method was established for the determination of levofloxacin in the human gastric mucosa.Thirty-seven patients with Helicobacter pylori (Hp) infection suffering from chronic gastritis participated in the study.All patients received therapy with 7 days of levofloxacin-based triple regime (esomeprazole 20 mg plus amoxicillin 1 000 mg,twice a day and levofloxacin 500 mg once a day).They received gastroscopy and were drawn three mucosal tissue from antrum after the end of treatment.The gastric mucosa were homogenized.The concentration of levofloxacin in gastric mucosa was determined by HPLC.A Hypersil ODS C18 column was used with the mobile phase of acetonitrile:50 mmol/L citric acid ∶ 1 mol/L ammonium acetate solution (27:72:1) at the detection wavelength of 295 nm.It was linear in the range of 0.5-10 μg/g.The recoveries for levofloxacin of low,medium and high concentrations were 96.4 %,95.2% and 102.8%.The intra-and inter-day RSDs were 6.70%,2.01%,1.46% and 7.40%,4.56%,3.62%,respectively.The concentration of levofloxacin in gastric mucosal were (6.05±2.47)μg/g in these patients.
