摘要
目的:探讨蛋白指纹图谱技术筛选宫颈癌患者血清中可用于诊断的特异性蛋白标志物。方法:采用弱阳离子纳米磁珠微球捕获的蛋白,用PBSⅡ-C型蛋白芯片阅读仪检测绘制成蛋白指纹图谱。所有蛋白指纹图谱采用Bomarker Wizard软件分析之后,用Biomarker Pattern软件识别最终用于诊断的蛋白标志物,并优化组合建立诊断模型。结果:在宫颈癌与对照组之间找到74个差异蛋白峰(P<0.01),其中质荷比(m/z)为3947 Da,4175 Da,5906 Da的蛋白峰组成的诊断模型可以将宫颈癌患者和健康志愿者区分,敏感性达94.0%(47/50),特异性为90.8%(59/65)。经双盲实验验证,该模型对宫颈癌诊断的敏感性达90.5%(38/42),特异性为96.0%(48/50)。结论:MALDI-TOF-MS技术联合纳米磁珠可以用来检测宫颈癌血清中的特异性蛋白标志物,并建立敏感性和特异性均较高的宫颈癌诊断模型。
Objectives:To screen the potential protein biomarkers in serum for the diagnosis of cervical cancer by protemic fingerprint.Methods:Proteomic fingerprint combining magnetic beads with MALDI-TOF-MS was used to profile and compare the serum proteins of patients with cervical cancer and healthy volunteers.PBSII-C protein chips reader was used to analyze protein peaks for significant difference.The protein fingerprint expression of all the serum samples and the resulting profiles between cancer patients and volunteers were analyzed with Bio-marker Wizard system.Results:Seventy-four differentially expressed proteins in serum were screened between pa-tients with cervical cancer and health individuals (P〈0.01).Proteins (3974 Da,4175 Da,5906 Da)were used for building a clinprot model which was able to classify patients with cervical cancer and health individuals with a sensitivity of 94.0%(47/50)and a specificity of 90.8%(59/65).Blind test data indicated a sensitivity of 90.5%(38/42)and a specificity of 96.0%(48/50).Conclusion:The biomarks in serum for cervical cancer can be dis-covered by MALDI-TOF-MS combining with magnetic beads,which is a powerful and reliable diagnostic method for cervical cancer with a high sensitivity and specificity.
