摘要
在大鼠成骨细胞体外培养过程中添加不同浓度的镉,利用xCELLigence细胞实时分析系统观察细胞指数、蛋白免疫印迹杂交检测核因子κB受体活化因子配体(RANKL)、骨保护素(OPG)蛋白表达,研究镉对成骨细胞毒性的影响。结果表明:镉暴露可使成骨细胞指数降低,0.5μmol·L-1镉对细胞增殖和存活具有抑制作用,呈剂量-效应和时间-效应关系;随着镉染毒剂量的增加,RANKL蛋白表达量降低(P<0.01);1.0μmol·L-1镉暴露组OPG蛋白表达量明显增加(P<0.05),镉浓度增大则导致OPG蛋白表达量降低(P<0.01或P<0.05);OPG/RANKL比值升高(P<0.01)。证明镉暴露通过RANKL/RANK/OPG系统调控骨代谢,效应可能与剂量有关。
A study on the effect of different concentrations of cadmium on rat osteoblasts (OBs) was carried out to estimate its toxic damage. Cell index were monitored by xCELLigence real-time analysis system. Also the expression of RANKL and OPG were measured by western blot. Results showed that, cadmium exposure decreased cell ifidex in a dose-effect and time- effect way; cell proliferation and survival could be inhibited by cadmium even with the dose of 0. 5 μmol · L-1 ; with the increase of cadmium concentrations, the expression of RANKL decreased (P〈0.01) while those of OPG increased signif- icantly (P〈0.05) in 1.0 μmol · L-1 cadmium group and then decreased (P〈0.01 or P〈0.05). Thus the ratio of OPG/ RANKL increased (P〈0.01). The study indicated that cadmium could regulate bone metabolism through RANKL/ RANK/OPG system, which may be related to its dosage.
