N-乙酰半胱氨酸对COPD大鼠模型血管活性物质的影响 被引量:5

Effects of N-acetylcysteine on Vasoactive Substances in Rats with Chronic Obstructive Pulmonary Disease

摘要 为观察N-乙酰半胱氨酸(NAC)抗氧化作用对慢性阻塞性肺疾病(COPD)模型大鼠血管活性物质的影响,将100只清洁级Wistar大鼠随机分为对照组、模型组、NAC低剂量组、中剂量、高剂量5组,每组20只。模型组采用气管注入脂多糖(LPS)和香烟染毒的复合方法造模。在模型组基础上,低剂量组、中剂量、高剂量组分别按26.32、52.64、105.28mg/kg NAC灌胃,连续2周。检测丙二醛(MDA)、超氧化物歧化酶(SOD)、血管紧张素Ⅱ(AngⅡ)、活性氧(ROS)、内皮素(ET)、一氧化氮(NO)及肺组织核转录因子κB(NF-κB)的变化,分析AngⅡ与ROS、NF-κB的相关性。结果显示,与对照组比较,模型组AngⅡ、ROS、ET、MDA水平均显著升高而NO、SOD水平极显著下降(P<0.01),NF-κB活性极显著升高(P<0.01)。与模型组比较,低剂量组NO水平升高而ET水平显著下降(P<0.05);中剂量组AngⅡ、ROS、ET、MDA水平下降或显著下降而NO、SOD水平均升高(P<0.05或P<0.01),NF-κB活性显著下降(P<0.05);高剂量组AngⅡ、ROS、ET、MDA水平均显著下降而NO、SOD水平均极显著升高(P<0.01),NF-κB活性极显著下降(P<0.01)。AngⅡ水平与NF-κB活性,AngⅡ水平与ROS水平均呈显著正相关关系(P<0.01)。表明NAC剂量依赖性改善COPD病理变化,推测其可能是通过降低AngⅡ水平,从而减少ROS的产生,进而抑制NF-κB活化过程而起效。 For the purpose of observing the effects of antioxidant N-acetylcysteine(NAC)on vasoactive sub-stances in rats with chronic obstructive pulmonary disease(COPD),100 Wistar rats,clear grade,were di-vided randomly into control group,model group,NAC low dose,middle dose and high dose 5 groups with 20 rats in each group.COPD model group was established with intratracheal instillation of lipopolysaccha-ride(LPS)and cigarette smoking.On the basis of mode group,the rats in low dose group,middle dose group and high dose group respectively were ig with NAC according to 26.32 mg/kg,52.64 mg/kg,105.28 mg/kg for 2 weeks.The levels of MDA,SOD,AngⅡ,ROS,ET,NO and NF-κB were examined,and s the corelation among AngⅡ,ROS and NF-κB was analyzed.Results showed that compared with the control group ,in the model group the levels of AngⅡ,ROS,ET and MDA all increased significantly(P 〈0.01) while the levels of both NO and SOD decreased significantly(P 〈 0.01),NF-κB activity increased(P 〈0.01).Compared with the model group,in the low dose group,the level of NO increased (P 〈0.05)while the level of ET decreased(P 〈0.05);in the middle dose group ,the levels of AngⅡ,ROS,ET and MDA all decreased while the levels of both NO and SOD increased(P 〈0.05 or P 〈0.01),NF-κB activity de-creased(P 〈0.05);in high dose group the levels of AngⅡ,ROS,ET and MDA all decreased significantly while the levels of both NO and SOD increased significantly(P 〈0.01),NF-κB activity decreased signifi-cantly (P 〈0.01).Meantime,the corelation between the level of AngⅡ ,NF-κB activity and the level of ROS had a significant corelation(P 〈0.01).So NAC could improve pathological changes of COPD dose-de-pendently and It was supposed to take an effect by reducing the level of AngⅡ,then decreasing the level of ROS to inhibit the activated process of NF-κB.
