摘要
以马血为试验材料,采用超声波与CMC-Na结合提取血红素,在单因素试验基础上,通过Box-Behnken设计和响应曲面法分析试验结果,建立数学模型,确定最佳提取条件。研究得出最佳工艺条件:超声功率为338W、超声时间为10min、去离子水的添加量是红细胞(RBC)体积的5倍、提取pH为5.5。此条件下,血红素产率达到最大值0.42g/50mL,与模型高度拟合。
Heme was extracted from horse blood, using ultrasound method combined with CMC-Na. Based on thesingle factor experiment, the Box-Behnken design and response surface method were used to set up themathematical model and determine the best extraction conditions.As a result,the optimum process conditions wereobtained as followingultrasonic power was 338W, ultrasonic time was 10min,deionized water content was 5 timesthe size of red blood cells (RBC), and extraction pH was 5.5. Under these conditions, the yield of heme reachedmaximum 0.42g/50mL which was in good agreement with the predicted value.
