摘要
目的探讨结肠癌组织中RAS相关区域家族1A(RAS association domain family,RASSF1A)蛋白的表达意义。方法收集60例结肠癌及40例正常结肠黏膜组织,采用Western blotting检测RASSF1A蛋白在结肠癌组织中的表达情况(正常结肠黏膜组织为对照);利用免疫组织化学染色方法观察结肠癌及正常结肠黏膜组织中RASSF1A蛋白的表达情况,并分析其临床意义。结果①Western blotting结果显示,RASSF1A蛋白在正常结肠黏膜中表达高于结肠癌组织。②免疫组织化学染色结果显示,RASSF1A蛋白在正常结肠黏膜中的表达阳性率为95.0%(38/40),在结肠癌中为46.7%(28/60),正常结肠组织中RASSF1A蛋白的表达阳性率明显高于结肠癌组织(P<0.01)。Duke’s分期的A+B期RASSF1A蛋白表达阳性率为61.1%(22/36),而C+D期为25.0%(6/24);有淋巴结转移者RASSF1A蛋白表达阳性率为11.1%(2/18),而无淋巴结转移者为61.9%(26/42),RAS SF1A蛋白表达与Duke’s分期和淋巴结转移有关(P<0.01)。结论正常结肠组织中RASSF1A蛋白表达明显高于结肠癌组织,结肠癌中RASSF1A蛋白表达与Duke’s分期和淋巴结转移有关。
[Objective] To investigate the expression of RASSF1A protein in colon cancer. [Methods] Werespectively collected 60 cases colon cancer and 40 cases normal colonic mucous membrane tissues. Mean-while, the expression of RASSF1A protein was investigated by Western b[otting in colon cancer, and normalcolonic mucous membrane was the control; Moreover, the expression of RASSF1A protein was observed by im-munohistostaining in colon cancer and normal colonic mucous membrane tissues, and the clinical significanceof RASSF1A protein expression was evaluated in different tissue. [Results] The Western blotting resultdisplayed that the expression of RASSF1A protein was higher in normal colonic mucous membrane tissuethan in colon cancer tissue. The immunohistostaining consequence exhibited that the positive expression ratesof RASSF1A protein were 95.0% (38/40) and 46.7% (28/60) in normal colonic mucous membrane and coloncancer tissues. The positive expression rate of RASSF1A protein was obviously higher in normal colonic mu-cous membrane tissue than in colon cancer tissue (P〈0.01). The positive expression rates of RASSF1A proteinwere 61.1% (22/36) and 25.0% (6/24) in colon cancer tissue of Duke's A+B stage and C+D stage; The posi-tive expression rates of RASSF1A protein were 11.1% (2/18) and 61.9% (26/42) in colon cancer tissue oflymphaden metastasis and non-lymphaden metastasis. The expression of RASSF1A protein was related withDuke's stage and lymphaden metastasis (P〈0.01). [Conclusion] The expression of RASSF1A was obviouslyhigher in normal colonic mucous membrane tissue than in colon cancer tissue and relevant with Duke's stageand lymphaden metastasis.
