血管生成素及血管内皮生长因子在进展期宫颈癌中的表达及其与放化疗相关性 被引量:6

Expression of angiopoietin and vascular endothelial growth factor in cervical cancer and influence of radiotherapy and chemotherapy

摘要 目的探讨血管生成素-1、血管生成素-2(Ang-1/2)和血管内皮生长因子(VEGF)在子宫颈鳞状细胞癌(SCC)组织中的表达及与放化疗相关性。方法入选49例单纯手术治疗的子宫颈鳞状细胞癌(SCC-O),25例术前经过化疗后再手术治疗者(SCC-CO)以及18例术前经过放疗再手术治疗的子宫颈鳞状细胞癌(SCC-RO)3组患者,用免疫组织化学染色法检测Ang-1、Ang-2及VEGF的表达,分析其与CD34标记的微血管密度(MVD)及D2-40标记的微淋巴管密度(LVD)的关系。结果从宫颈癌Ⅰ期到Ⅱ期Ang-2蛋白及VEGF蛋白表达强度有所增强,而Ang-1蛋白的表达则下降;放、化疗使Ang-2及VEGF蛋白表达强度明显降低(P<0.05),且使MVD值及LVD值显著降低(P<0.05);Ang-2及VEGF表达强度与MVD值及LVD值呈显著正相关(P<0.05)。结论 Ang-2及VEGF在子宫颈癌中的高表达与肿瘤进展有关;Ang-2与VEGF在促进宫颈癌血管及淋巴管的形成上起相互协同作用;放疗及化疗可明显抑制宫颈癌Ang-2与VEGF表达和血管及淋巴管的形成。 Objective To investigate the expression of angiopoietin -1 ( Ang - 1 ), angiopoietin - 2 ( Ang - 2) and vascular endothelial growth factor (VEGF) in cervical carcinoma (SCC) tissues and relations with radiotherapy and chemotherapy. Methods Immunohistochemistry SP method was used to detect the expression of Ang - 1, 2 and VEGF in 3 groups : 49 cases of cervical squamous cell carcinoma ( SCC - O) treated with simple treatment of operation, 25 subjects with chemotherapy treatment before operation( SCC -CO) and 18 cases of cervical squamous cell carcinoma treated with preoperative radiotherapy and followed operation (SCC - RO ). And the relations of those expressions with microvessel density CD34 positive markers (MVD) and D2 -40 marker- positive lymphatic vessel density (LVD) were analyzed. Results The expression Ang - 2 and VEGF were increased from stage Ⅰ to Ⅱ of cervical cancer( P 〉 0.05 ) , and the expression of Ang - 1 protein was in the opposite. Radiotherapy and chemotherapy obviously decreased the expression of Ang- 2 and VEGF (P 〈 0. 05), as well as both the MVD value and LVD value (P 〈 0. 05 ). The expression of Ang - 2 and VEGF were positively correlated with the value of MVD and LVD in the tumor(P 〈 0.05). Conclusion High expressions of Ang - 1, Ang - 2 and VEGF in cervical cancer tissue might play an important role in the tumor advancement. The expressions of Ang -2 and VEGF are synergic to improve generation of cervical blood and lymphatic vessels. Cervical cancer radiotherapy and chemotherapy could significantly inhibit the formation of blood vessels and lymphatic vessels in tumor tissue.
