摘要
目的:研究人脐带血间充质干细胞(HUCB-MSC)对人胃癌MGC80-3细胞体外细胞外基质成分的影响,探讨其对胃癌细胞生物学行为的影响。方法:原代培养人脐带血间充质干细胞,流式细胞仪进行细胞表型鉴定。采用流式细胞仪半定量检测基质金属蛋白酶-7(MMP-7)及基质金属蛋白酶抑制剂-1(TIMP-1)蛋白水平。逆转录-聚合酶链反应半定量测定MMP-7和TIMP-1的表达情况。结果:各实验组中MMP-7表达的蛋白荧光指数(FI)均低于空白对照组,各实验组中TIMP-1表达的蛋白荧光指数(FI)均高于空白对照组。MMP-7表达的分子水平随MSC-CM浓度增加而逐渐降低,TIMP-1表达的分子水平随MSC-CM浓度增加而逐渐增高。各实验组与空白对照组比较均有统计学意义(P<0.05)。结论:MSC-CM可下调MMP-7、上调TIMP-1,这可能是HUCB-MSC抑制胃癌MGC80-3细胞迁移能力的机制之一。
Objective:To study the effects of human umbilical cord blood mesenchymal stem cells on extracellular matrixc of gastric carcinoma cells MGC80-3,to investigate its effects on cell biological behaviors of gastric carcino-ma. Methods:HUCB-MSCs were primary cultured in vitro and identified the phenotype. The protein level of MMP-7 and TIMP-1 were semi-quantitatively detected by flow cytometry. The mRNA level of MMP-7 and TIMP-1 were semi-quantitatively detected by reverse transcription PCR. Results:The expression of MMP-7 protein fluores-cence index( FI)was decreased with the increase of MSC-CM concentration. The expression of TIMP-1 protein fluorescence index was increased with the increase of MSC-CM concentration. The expression level of MMP -7, TIMP-1 in different experimental groups had significant statistical difference. The expression level of MMP-7 was decreased with the increase of MSC-CM concentration. The expression level of TIMP-1 was increased with the in-crease of MSC-CM concentration. There were significant differences between experimental groups and control group (P〈0. 05). Conclusion:MSC-CM could inhibit the proliferation of human gastric cancer cell MGC80-3 through inhibiting the level of MMP-7 and upregulating the level of TIMP-1.
