反复丙泊酚麻醉对大鼠学习记忆功能的影响 被引量:10

摘要 目的探讨反复使用不同剂量的丙泊酚麻醉,对成年SD大鼠的空间学习记忆功能的影响。方法 8周龄雄性SD大鼠32只,随机分为4组:镇静剂量丙泊酚组(Prop 5)、低剂量丙泊酚组(Prop 20)、高剂量丙泊酚组(Prop 30)和10%脂肪乳剂组(Lip)。丙泊酚的3个剂量组,首先均采用15 mg/kg的丙泊酚作为诱导剂量。Prop 5组、Prop 20组和Prop 30组分别以5、20、30 mg/(kg·h)的剂量维持麻醉2 h;Lip组首先给予15 mg/kg的脂肪乳剂,之后脂肪乳剂维持输液量与Prop 30等毫升数。所有药物均采用大鼠尾静脉注射,每日1次,连续5 d。第6天起,选用Morris水迷宫进行行为学实验。行为学实验结束后,立即取大鼠海马组织,行H-E及Bax抗体免疫组织化学法染色,并观察海马组织超微结构的变化。结果行为学实验中,3组不同剂量丙泊酚组寻找平台潜伏期、首次穿越平台时间均明显延长(P<0.05),穿越平台次数也减少(P<0.05)。海马组织H-E染色无明显异常;Bax抗体实验显示,Prop 20组及Prop 30组染色显现棕黄色凋亡细胞颗粒。电子显微镜下,Prop 20组丙泊酚的海马神经元细胞肿胀,Prop 30组神经细胞内产生自噬现象。结论在本实验条件下,成年SD大鼠接受不同剂量丙泊酚的反复麻醉后,抑制其空间学习记忆能力,海马组织超微结构发生改变。 Objective To investigate the effects of repeated use of propofol on learning and memory function in rats. Methods Thirty tw o 8-w eek-old male Sprague Daw ley( SD) rats w ere randomly assigned to sedative propofol group( Prop 5),low dose propofol group( Prop 20),high dose propofol group( Prop 30) and 10% intralipid group( Lip) w ith 8 in each group. The rats in propofol groups w ere given w ith 15 mg / kg propofol intravenously as induction dose,then w ith 5 mg /( kg · h), 20 mg /( kg·h) or 30 mg /( kg ·h) propofol respectively for 2 h. Meanw hile,the rats in Lip group w ere given 15 mg / kg intralipid intravenously first,then intralipid w as continuously infused w ith the volume as used in Prop 30 group. All the drugs w ere given once a day,lasting for 5 d. Learning and memory function of rats w as tested w ith place navigation and spatial probe tests in Morris w ater maze. After the animals w ere sacrificed the sections of hippocampus w ere prepared for the H-E and immunohistochemical staining; and the ultrastructure of hippocampus w as observed w ith transmission electron microscopy( TEM). Results In propofol groups,the latency w as significantly longer( P &lt; 0. 05) compared to the Lip group. Immunohistochemical staining show ed apoptotic cell granules in hippocampus and TEM also demonstrated noticeable changes in hippocampal neurons of rats in Prop20 and Prop30 groups. Conclusion Repeated anesthesia w ith propofol affects spatial learning and memory function of the Sprague Daw lery rats.
