摘要
从土壤中筛选得到1株产胞外葡萄糖氧化酶的黑曲霉菌株1504,通过诱变育种选育得到突变菌株UNⅡ021。为进一步提高产酶活力,对其培养基成分和培养条件进行优化,结果最佳培养基组成和培养条件为:以10%的葡萄糖为碳源,0.5%牛肉蛋白胨和0.5%硝酸钠为氮源,碳酸钙添加量30 g/L,培养基初始pH 5.2,接种量0.05(V/V),培养温度29℃,摇床转速240 r/min。在此条件下,培养4 d菌株UNⅡ021产葡萄糖氧化酶活力达到363.04 U/mL,是优化前的2.14倍。
A strain producing extracellular glucose oxidase named Aspergillus Niger 1504 was screened from soilsamples and its mutant UN 11 021 was obtained by mutation breeding. Higher activity of extracellular glucose oxidase wasobtained optimizing the medium composition and culture conditions with the best conditions as followed 10% glucose ascarbon source, 0.5% beef extract peptone and 0.5% sodium nitrate as nitrogen source, the amount of calcium carbonate30g/L, inoculation amount 0.05 (V/V). The optimum temperature and pH were 29 ℃ and 5.2 respectively, with shakingspeed was 240 r/min. The production of extracellular glucose oxidase by UN 11 021 could be 363.04 U/mL by the end ofthe Forth day, which was 2.14 times more than the original one.
