摘要
目的研究人参皂苷对体外纯化培养大鼠视网膜Müller细胞的影响,为人参治疗糖尿病性视网膜病变(DR)的物质基础研究筛选合适的浓度。方法用不同浓度的人参皂苷提取物及人参皂苷Re、人参皂苷Rg1、人参皂苷Rb1对照品分别干预体外纯化培养的大鼠视网膜Müller细胞48 h,MTT法检测细胞增殖抑制率。结果测得人参皂苷提取物及人参皂苷Re、Rg1、Rb1对照品的IC50分别为0.618,0.489,0.653,0.553 mg/ml,给药浓度为1 mg/ml时,测得最高细胞增殖抑制率分别为56.8%,71.0%,59.4%,68.0%。结论人参皂苷提取物及人参皂苷Re、Rg1、Rb1对照品对大鼠视网膜Müller细胞均有一定抑制作用,且具有剂量依赖性,所测IC50值为进一步研究人参皂苷提取物治疗DR作用机制的有效浓度提供依据。
Objective To study the effect of ginsenoside extractions on rat retinal Mtiller cell, screening appropriate concentration for diabetic retinopathy(DR) substances basic research. Methods The measured different concentration of ginsenoside extractions and ginsenoside Re,ginsenoside Rgl,ginsenoside Rbl control products were applied to rat retinal Mtiller cells 48h, MTT method was used to detect the toxic effect on cells. Results The measured ICs0 of ginsenoside extractions and ginsenoside Re, Rgl, Rblwere 0. 618 mg/ml,0. 489 mg/m1,0. 653 mg/m1,0.553 mg/ml ,to the concentration of 1 mg/ml ,the measured maximum inhi- bition rates were 56.8% ,71.0% ,59.4% ,68.0%. Conclusion The ginsenoside extractions and ginsenoside Re, Rgl ,Rbl on rat retinal Mtiller cells have a certain inhibition, with the concentration increased, the inhibition increased. The IC50 values provide the basis for further research on the DR mechanism of effective concentration.
