人参三醇组皂甙对人视网膜色素上皮细胞增生和DNA合成的抑制作用 被引量:4

摘要 目的探讨西洋参茎根源人参三醇组皂甙(Panoxatriol saponins,PTS)对体外培养视网膜色素上皮细胞(RPE)增生的抑制作用及可能机制。方法通过活细胞计数法、MTT比色法和3H-TdR掺入法分别检测不同浓度(500、4003、00、2001、005、01、0、5μg/ml)的PTS以及PTS200μg/ml在不同作用时间(12-120小时)对RPE细胞增生和代谢的影响。结果 PTS组细胞增殖受抑并出现细胞脱落,PTS400μg/ml具有最强的抑制效应,其明显抑制效应在12小时出现,96小时达高峰。PTS组A植和cpm值明显低于对照组,PTS的EC50为64.3μg/ml。RPE细胞生存率和3H-TdP掺入率下降。结论 PTS可能通过抑制DNA合成、干扰RPE细胞代谢对RPE细胞增殖具有剂量依赖和时间依赖方式的抑制作用。 Objective To study the inhibition on RPE in vitro by PTS and to investigate the possible mechanism.Methods We tested the effect on RPE proliferation and metabolism by using different concentration(500,400,300,200,100,50,10,5 μg/ml) PTS via cytometry,MTT assay and 3H-TdR incorporation.We also tested different cultural time(12-120 hours) when PTS concentration is 200 μg/ml.Results In PTS group,the cells started falling off from the Petri dish and the proliferation was inhibited.The 400 μg/ml PTS had best inhibition.It worked in 12 hours obviously and reached its peak at 96 hours.A value and cpm value were lower than the control group.EC50 was 64.3 μg/ml.The RPE survival rate and 3H-TdR incorporation decreased.Conclusion PTS can inhibit the RPE proliferation possibly by inhibiting DNA synthesis and intervening RPE metabolism.The effect was dose and time dependent.
