摘要
目的观察下肢缺血再灌注后糖尿病大鼠血清、坐骨神经组织中细胞间黏附分子-1(ICAM-1)和髓过氧化物酶(MPO)的表达情况,探讨下肢缺血再灌注对糖尿病大鼠坐骨神经的损伤机制。方法 24只清洁级SD雄性大鼠,分为正常组(n=6)和糖尿病组(n=18),糖尿病组采用链脲佐菌素造模,饲养1个月后,选择部分糖尿病大鼠(n=9)作为糖尿病缺血再灌注组,暂时阻断腹主动脉、髂总动脉和股动脉3h,再灌注7天后检测各组大鼠血清、坐骨神经中ICAM-1和MPO的表达情况。结果糖尿病缺血再灌注大鼠血清、坐骨神经组织中ICAM-1和MPO含量明显高于糖尿病大鼠和正常大鼠(P<0.01)。结论糖尿病大鼠下肢缺血再灌注通过提高血清、坐骨神经组织中ICAM-1和MPO的表达水平参与坐骨神经的损伤过程。
Objective To observe the expression level of intercellular adhesion molecule - 1 (ICAM - 1 ) and myeloperoxidase (MPO) in serum and sciatic nerve tissue after ischemia- reperfusion, and study how the isehemia -reperfusion of diabetic rats' lower limb injures sciatic nerve. Methods Twenty - four male SD rats were divided into two groups: normal control group ( n = 6) , diabetic group( n = 18) , and the model rats suffering from diabetes were induced by STZ. After feeding for one month, some of the model rats suf- fering from diabetes (n = 9)were chosen as the isehemia - reperfusion group and kept abdominal aorta, common iliac artery and femoral ar- tery blocked for 3 hours. After 7 - day perfusion, the expression level of ICAM - 1 and MPO in all rats' serum and sciatic nerve tissue was detected. Results ICAM - 1 and MPO of serum and sciatic nerve in diabetic rats suffering from ischemia - reperfusion are higher obvi- ously than those from normal control group and diabetic group without isehemia - reperfusion (P 〈 0.01 ). Conclusion Ischemia - reperfusion in diabetic rats' lower limb can participate in the injury process of sciatic nerve by improving the expression level of ICAM - 1 and MPO in serum and sciatic nerve tissue.
