摘要
岩藻黄素是存在于褐藻中的1种类胡萝卜素,具有多种生物活性。本文用福建省生产的干海带为原料,研究海带中岩藻黄素浸提的最佳有机溶剂及工艺优化参数。以Sigma标样作为参照下,对经两次硅胶层析柱分离纯化的岩藻黄素样品进行液相色谱(HPLC)定量分析。试验结果表明,所选7种溶剂中甲醇提取效果最好,在60℃,料液比为1∶40(g/mL)的条件下避光静置,浸提2次,每次1 h,岩藻黄素的提取率达到0.375 mg/g。经HPLC鉴定,岩藻黄素的纯度约为86%。本研究结果为高效利用海带原料及其下脚料提取岩藻黄素活性物质提供技术基础。
Fucoxanthin,widely and largely existing in brown algae,is one of carotenoid which has a variety of biological activities.In this work,the dried Laminaria japonica produced in Fujian was selected as the experimental material for the high content of fucoxanthin.The best extracting solvent and related technological parameters in the processes of extracting fucoxanthin were need to optimize.Subsequently,In the reference of fucoidan standard samples(Sigma).Then the fucoxanthin samples which was purified and separated twice times by silica gel column chromatography were detected by high-performance liquid chromatography(HPLC).The results showed that methanol is the most suitable organic solvent for extracting fucoxanthin among the seven kinds of selected organic solvents,and the best extracting processes for fucoxanthin as follows: extracting temperature,60 ℃; the ratio of material and extracting solvent,1∶40(W/V,g/mL); extracting time,2 times and each time 1 h.In this condition,the best extracting rate of fucoxanthin was 0.375 mg/g.After purified,the purity of fucoxanthin was about 86%.It is indicated that silica gel column chromatography is efficient for purifying fucoxanthin.This work has provided theoretical basis for efficiently utilizing the scraps of Laminaria japonica or Laminaria japonica to obtain fucoxanthin.
