养殖多肋藻生长期营养成分测定与分析 被引量:4

Variations in Nutritional Composition in Cultivated Seaweed Costaria costata during Growth Period

摘要 对山东荣成楮岛湾海域养殖多肋藻3—6月生长期的营养成分进行了跟踪测定。试验结果显示,多肋藻基本营养成分以灰分(23.29%~30.44%)、褐藻胶(25.95%~27.86%)为主,其次是甘露醇(15.42%~16.64%)和粗蛋白(7.44%~11.43%)。灰分和粗蛋白在3—6月呈降低趋势,而甘露醇和褐藻胶无明显变化。利用反相高效液相色谱法测定多肋藻藻粉中主要色素含量,发现叶绿素a(15.05~330.30μg/g)和岩藻黄素(42.07~159.93μg/g)均随生长快速降低,β-胡萝卜素含量低(1.72~8.88μg/g)。采用热水浸提,乙醇分级沉淀的方法制备褐藻糖胶粗品,利用高效液相色谱法分析粗品中褐藻糖胶的单糖组成,硫酸钡比浊法测定硫酸基含量,结果显示,褐藻糖胶含量达到藻粉干质量的1.79%~2.10%,其中硫酸基占褐藻糖胶的38.57%~47.26%。所有样品均含有甘露糖、氨基葡萄糖、鼠李糖、葡萄糖醛酸、葡萄糖、半乳糖、木糖、岩藻糖8种单糖。其中岩藻糖为主要单糖,其摩尔百分含量(40.11%~59.89%)在3—6月份呈不断增长趋势;其次为半乳糖(22.54%~34.73%),变化趋势与岩藻糖相反;其他6种单糖摩尔百分含量较低,除木糖外,各单糖的月份差异明显。 The nutritional compositions were monthly analyzed in cultivated seaweed Costaria costata collected from Chudao Bay,Rongcheng,Shandong Province during growth period(from March to June in 2017)to investigate the variations in nutrition of the seaweed.The results showed that the proximate composition of the seaweed was dominated by ash(23.29%—30.44%)and alginate(25.95%—27.86%),followed by mannitol(15.42%—16.64%)and crude protein(7.44%—11.43%).The contents of ash and crude protein were found to be decreased with the growth of the seaweed,without significant fluctuation in contents of alginate and mannitol during growth period.Reversed-phase high performance liquid chromatography(HPLC)analysis indicated that contents of both chlorophyll a(15.05—330.30μg/g)and fucoxanthin(42.07—159.93μg/g)were rapidly decreased with growth,with much lowerβ-carotene content than other two pigments(1.72—8.88μg/g)in the seaweed.There was 1.79%—2.10%of fucoidan in dried seaweed,sulfate content accounting for 38.57%—47.26%of fucoidan,and the monosaccharide composition of crude fucoidan was analyzed by HPLC.HPLC analysis revealed that the seaweed contained mannose,glucosamine,rhamnose,glucuronic acid,glucose,galactose,xylose and fucose,in which fucose was the main monosaccharide,accounting for 40.11%—59.89%,showing an increase trend from March to June.Galactose was the second compound(22.54%—34.73%)in content with an opposite trend to fucose,and the remaining monosaccharides being lower in content,were affected significantly by growth with an exception of xylose.The findings indicate that seaweed C.costata is an excellent resource of bioactivity constituents in the commercial utilization.
