摘要
目的:研究仙灵复方制剂经灌胃给药后对去势大鼠雌激素水平、骨密度和骨形态的影响。方法:雌性6月龄SD大鼠分为假去势组、模型组、阳性对照组、仙灵复方制剂高、中、低剂量组共6组(n=12),除假去势组进行假手术外,其余均采用彻底摘除卵巢法制造实验型骨质疏松模型,术后进行模型筛选,灌胃给药,连续给药3个月。末次给药后,检测一些生化指标并测定骨密度、骨生物力学和骨组织形态。结果:与模型组比较,仙灵复方制剂组在大鼠血清中Ca2+、P、SOD、E2含量显著增高(P<0.05),MDA的含量显著降低(P<0.05),增加腰椎骨密度,提高模型大鼠的股骨最大载荷和刚度,改善骨小梁数量和连续性。结论:仙灵复方制剂可明显改善去卵巢大鼠的骨生物力学性能,提高去势大鼠血清中雌激素水平,能防治去卵巢诱导的大鼠骨质疏松症,是值得开发的一种中药复方制剂。
Objectives:To study the effects of XianLing FuFang Preparation on osteoporosis in ovariectomized rats. Methods:The female 6-month-old SD rats were divided into sham operated group, ovariectomized group, high concentration XianLing FuFang Preparation with ovariectomized group, medium concentration XianLing FuFang Preparation with ovariectomized group and low concentration XianLing FuFang Preparation with ovariectomized group. Some biochemical indexes were examined after three months of administration. Bone mineral density (BMD) of lumbar vertebra bone of rats was determined by Dual Energy X-ray Absorptiometry(DEXA). Biomeehanieal characteristics of femur were measured by three point bending test. some tissue from the right thighbone were undertaken morphologic observation. Results :The levels of calcium and phosphorus in serum were significantly higher in the rats treated by different dosages of XianLing FuFang Preparation as compared with that in the osteoporosis control group. As for the level of E2, Ca2+ , P, SOD and MDA in serum, the difference was significant (P 〈 0. 05) between the treated group and the control group. The resistanee of bending and compression of femur of rats was enhanced, and the bone trabecula numbers and continuity of XianLing FuFang Preparation groups are obviously better than the other group. Conclusion: XianLing FuFang Preparation had antiosteoporotic effects on ovariectomized rats, and it was worth promoting.
