摘要
研究以纯化猪皮为原料,采用酸-酶结合法制备得到了I型胶原。再以市售高脱乙酰度壳聚糖为原料,采用乙酰化反应,制备一定乙酰度的壳聚糖。接着将所提取的I型胶原与壳聚糖以不同质量比例混合制备复合海绵,并使用红外、DSC、AFM等对自制壳聚糖和复合海绵的结构进行表征,检测了壳聚糖的特性黏度。结果表明:红外证实了经乙酰化反应后的市售壳聚糖的酰胺键被有效置换,制备得到的壳聚糖的特性黏度为825.584,粘均相对分子质量为2.43×105;复合后I型胶原的三股螺旋结构存在一定程度地变化,随着壳聚糖含量的增加,复合海绵的热稳定性提高,有利于样品的存放。
The collagen was extracted from the purified pig skin by the acid swelling-pepsin digestion process, and then the designed chitosan was also preparedusing commercial high degree deacetylatedof chitosans as raw material. Finally, the compound collagen sponge was manufactured by mixing collagenwith deacetylated chitosan under different mass ratio. After that the structure of the compound collagen sponge wascharacterized by SDS-PAGE, FT-IR spectra, Differential Scanning Calorimeter (DSC) and Atomic Force Microscope (AFM). FI-IR spectra of chitosan showed that the amido bond can be replaced effectively. The intrinsic viscosity ofhomemade is 825.584. The viscosity-average molecular weight is 2.43×10^5. The triple helix structure of compound collagen suffer certain damage. The thermal stability of compound sponge increase with the content of chitosan increase, which can enhance the maintain property of the collagen sponge.
